With the motivation to expand the compound library of specifically substituted pyrimidine derivatives, we prepared several pyrimidine-4-carbaldehydes. In most cases, the chemoselective oxidation of 4-hydroxymethyl-substituted pyrimidine derivatives could be achieved in good yields to provide the desired compounds. Alternatively, the aldehydes were prepared by Riley oxidation with selenium dioxide from the corresponding 4-methylpyrimidines. The formyl group of these compounds was employed as handle to generate alkynyl, cyano, oxazol-5-yl or β-ketoester substituents. Furthermore, two methods were found to prepare furo[3,2-d]pyrimidines. A serendipitously discovered reaction to a 6-(pyrimidin-4-yl)furo[3,2-d]pyrimidin-7-ol derivative involves a mechanistically interestingly ‘dimerization’ process with a benzoin addition as key step. Other compounds in this series contain amino, azido, or 1,2,3-triazol-1-yl groups. All these transformations to highly substituted pyrimidine derivatives demonstrate the synthetic versatility of pyrimidine-4-carbaldehydes and similar compounds.

Sustainable green heterocycles compounds of fused bi, tri, and tetracyclic systems containing pyrimidine-barbituric acid rings were synthesized via Hantzch reaction in a simple and efficient method using one-pot three-component of bis aldehydes, barbituric acid, and substituted amines the reactions occurred under microwave irradiation and in catalyst-free conditions. The products were produced with high yields and simple workups. This article emphasizes the significance of sustainable green philosophy that applies to synthesis methods for fused pyrimido[4,5-d]pyrimidine systems, highlighting their environmental friendliness advantages, efficiency, and safety.

Sustainable Green Synthesis and Molecular Docking Study of a Bis-Fused System Incorporating Pyrido[2,3-d]pyrimidine using Nanocatalyst under Microwave Condition

Aims: This study aimed to employ sustainable green methods in the synthesis of bisfused cycles incorporating pyrido[2,3-d]pyrimidine moiety using a green catalyst nano ZnO catalyst by one-pot, multicomponent reaction among 2,2'-(propane-1,3-diylbis(sulfanediyl)) bis(6-aminopyrimidin-4(3H)-one) 3, 1H-indene-1,3(2H)-dione 4, and aromatic aldehydes 5. Method: The reactions proceeded with both conventional and microwave (MW) irradiation methods. Result:: The microwave-assisted method carried out the reaction in 10 min and had high yields (89-95%). Conclusion: A molecular docking simulation study was conducted using human serum albumin (PDB: ID (2XVQ)). The study revealed that compounds strongly fit into the active sites of the target protein.
