Synergistic Antitumor Activity of Sorafenib in Combination with Epidermal Growth Factor Receptor Inhibitors in Colorectal and Lung Cancer Cells

Abstract: Purpose: Cancer cell survival, invasion, and metastasis depend on cancer cell proliferation and on tumorinduced angiogenesis. We evaluated the efficacy of the combination of sorafenib and erlotinib or cetuximab.Experimental Design: Sorafenib, erlotinib, and cetuximab, alone or in combination, were tested in vitro in a panel of non-small cell lung cancer (NSCLC) and colorectal cancer cell lines and in vivo in H1299 tumor xenografts. Results: Epidermal growth factor receptor (EGFR) ligand mRNAs were expressed in … Show more

“…Specifically, this multi-TKI exhibited cytotoxic effects in different CRC cell lines regardless of their KRAS status, as described in other preclinical studies (23,24) and suggested by several phase I studies, involving a large number of patients with CRC (25)(26)(27)(28).…”

“…GEO and SW48 cells are differentiated colon carcinoma cells with little or no ability to migrate or to invade in vitro (35). GEO-CR and SW48-CR cells acquired the ability to migrate in an in vitro migration assay ( Fig.…”

“…Cross-talk between the EGFR-dependent autocrine pathway and of VEGFR-dependent signaling in cancer cells has been shown by the study of Martinelli et al (2010). Moreover, they evaluated the in vitro and in vivo antitumor activity of anti-EGFR drugs, such as erlotinib, cetuximab and sorafenib.…”

“…Moreover, they evaluated the in vitro and in vivo antitumor activity of anti-EGFR drugs, such as erlotinib, cetuximab and sorafenib. Martinelli et al (2010) displayed the anti-proliferative effects of sorafenib on non-small cell lung cancer and colorectal cells (Martinelli et al, 2010). Simonelli et al (2013) used sorafenib and lapatinib combination in 30 patients with refractory solid tumors and reported that combination of sorafenib and lapatinib achieved in stabilization of disease.…”
