Synergistic binding of hydrophobic probes and zinc ligands to thermolysin

Abstract: The strong synergism previously observed in the binding of inhibitors to two Zn-proteases, has also been found for thermolysin. As in earlier cases, the effects are produced by a small Zn-ligand (e.g. a hydroxamate) in the presence of another compound which contains the key structural features of specific substrates (a specificity probe). For thermolysin, the most effective specificity probes are hydrophobic derivatives of amines and amino acids (e.g. carbobenzyloxy-L-alaninol). Even the simple combination of … Show more

“…The results reported in this study, taken with our previous work on three other Zn-proteases (DiGregorio et al, 1988;Pfuetzner and Chan, 1988;Pfuetzner and Chan, 1993) show a striking pattern of behaviour in this class of enzymes. Thus in all cases examined, synergistic binding to the enzyme requires the presence of a Zn-ligand and a probe with structural characteristics of specific substrates.…”

“…In the case of weak inhibitors or compounds with low solubility, the K, values were often determined more conveniently from the Yonetani-Theorell plot. The procedure is described in the preceding paper (Pfuetzner and Chan, 1993).…”

“…To investigate the possible existence of such interactions between subsites we have examined whether two inhibitors binding to different locations could exert effects on each other. In the past few years our laboratory has investigated the phenomenon of synergism in the binding of inhibitors to several zinc proteases [13][14][15] including the clinically important angiotensinconverting enzyme [16]. Although inhibitor synergism has not been reported in aspartate proteases, theoretical considerations suggest that it should be observable in a variety of enzymes [15].…”
