Synergistic effects of myo‐inositol and melatonin on cryopreservation of goat spermatozoa

Abstract: Overproduction of reactive oxygen species (ROS) during sperm cryopreservation has a detrimental effect on sperm parameters. Therefore, the use of antioxidants in the sperm freezing extender can reduce ROS destructive effects. In this study, we investigated whether co-supplementation of melatonin and myo-inositol into the semen extender can improve the post-cryopreservation quality of goat spermatozoa. After the freeze-thawing process, sperm motility, viability, plasma membrane and acrosome intact morphology we… Show more

“…Moreover, 5 mM oxidized glutathione increased the motion and velocity characteristics of ram sperm after freeze–thaw [ 92 ], and similar results were found at lower concentrations in turkey sperm [ 93 ]. Similar results have been found using the multifunctional antioxidant melatonin, where positive effects for sperm cryopreservation were found in pig [ 94 ], ram [ 95 ], goat [ 96 ], canine [ 97 ], fish [ 98 ], bovine [ 99 ], and human [ 100 ].…”

“…Melatonin, a type of indoleamine and free radical scavenger, has been reported to exhibit various antioxidative effects through G-protein-coupled receptors (GPCR). Recently reported studies have focused on the protective effect of melatonin in maintaining fertility in several species of animals and humans [ 25 , 27 , 28 , 29 , 30 , 31 , 34 , 39 , 137 , 138 , 139 ]. Even in humans, melatonin has been reported to be very useful as a sperm cryoprotectant and it has a significant effect on the toxicity and damage recovery of frozen sperm.…”

“…In the ram, melatonin inhibits the mitochondrial permeability transition pore opening by MT1-related PI3K/AKT/GSK3b signal activation [ 34 ]. Additionally, melatonin and Myo-inositol mixture have a critical effect on sperm activation during cryopreservation in goat sperm [ 39 ]. In pigs, a melatonin and butylhydroxytoluene mixture successfully reduced reactive nitrogen species and oxidative stress during a sperm vitrification procedure [ 36 ].…”

“…9 However, the excessive production of ROS due to the freeze-thawing process cannot be overcome by endogenous antioxidants owing to the limitations of the spermatozoa cytoplasm and decreased antioxidant levels due to the addition of extenders. 10 Several studies to improve the quality of post-thawing goats semen have been carried out, including adding extenders with egg yolk omega-3, 11 fish semen plasma, 12,13 curcumin, 14 butylated hydroxytoluene, 15 combination of myo-inositol and melatonin, 16 and L-carnitine. 17 Epigallocatechin-3-gallate (EGCG) is a powerful antioxidant extracted from green tea.…”
