Synergy between myogenic and non-myogenic cells in a 3D tissue-engineered craniofacial skeletal muscle construct

Abstract: In vitro skeletal muscle engineering involves the culture of isolated primary myogenic cells in an environment conducive to the formation of a three-dimensional (3D) tissue construct capable of generating force. Isolated human myogenic cells have been used to study cell-cell interactions, permitting identification of functions intrinsic to skeletal muscle in two dimensions (2D). However, the independent contribution of human myogenic and non-myogenic cell types that comprise skeletal muscle to myogenic cell di… Show more

“…The protocol for the construction of MDC populated collagen constructs was adapted from those detailed elsewhere [Brady et al, 2008;Mudera et al, 2010]. Briefly, 300 ml 10Â MEM (GIBCO/ Invitrogen) solution was added to 2.6 ml type 1 rat-tail collagen (First Link, Birmingham, UK) (in 0.1 M acetic acid, protein concentration n ¼ 2.035 mg/ml) and mixed thoroughly.…”

“…We have now further developed the model, removing it from the custom CFM system into standardized and commercially available chamber slides. Previous work using the CFM system has focused on the use of human derived muscle cells [Brady et al, 2008;Mudera et al, 2010] and the C2C12 mouse muscle cell line [Cheema et al, 2003]. In order to expand the application of the 3D collagen model as a whole, we have now developed optimum conditions for promoting the development and maturation of primary rat muscle cells.…”

“…Also, satellite cells harvested from the soleus muscle of rats are used for TE of muscle tissue (Dennis and Kosnik, 2000;Beier et al, 2004;Stern-Straeter et al, 2005;Das et al, 2006;Borschel et al, 2006;Larkin et al, 2006;Bach et al, 2006;Huang et al, 2006b;Boontheekul et al, 2007). Other muscles harvested for TE research include the latissimus dorsi and rectus femoris of rats (Kamelger et al, 2004), the flexor digitorum brevis of rats (De Coppi et al, 2005), the tibialis anterior of rats (Dennis et al, 2001;Huang et al, 2005;Boontheekul et al, 2007), the extensor digitorum longus of mice (Dennis et al, 2001), the human masseter (Lewis et al, 2000;Sinanan et al, 2004;Shah et al, 2005;Stern-Straeter et al, 2008;Brady et al, 2008a) and brachioradialis (Alessandri et al, 2004) and the iliofibularis from female Xenopus laevis frogs (Jaspers et al, 2006) (Table 2).…”

“…Sinanan et al (2004) investigated an approach to purify the population of myogenic satellite cells through CD56 + , which is a cell adhesion molecule (NCAM) expressed in proliferating myoblasts (Illa et al, 1992). Although the resulting cell population has a higher percentage of desmin (Sinanan et al, 2004), which is an intermediate filament protein, force production of this population was significantly lower than in the mixed and CD56 − cell populations (Brady et al, 2008b).…”
