Structurally diverse spiroheterocycles; spiro[pyrimido[2,1‐b]benzothiazole‐3,3′‐chromene]‐2′,4′‐dione, spiro[pyrimido[2,1‐b]benzothiazole‐3,5′‐pyrimidine]‐2′,4′,6′‐trione, and spiro[pyrimido[2,1‐b]benz‐thiazole‐3,2′‐cyclohexane]‐1′,3′‐dione have been synthesized by an environmentally benign, efficient, and facile one‐pot pseudo‐four component reaction of 2‐aminobenzothiazoles with aromatic aldehydes and cyclic β‐diketones in aqueous medium. The process involves hetero‐Diels–Alder cycloaddition and provides facile access to spiroheterocycles fused with potentially interesting biologically active scaffolds. The configuration of hetero‐Diels–Alder cycloadduct has been ascertained through density functional theory calculations.

Synthesis and in vitro antiproliferative activity of novel 12(H)-quino[3,4-b][1,4]benzothiazine derivatives

Novel method of N-dealkylating quinobenzothiazinium salts 2, promoted by reaction with benzimidazole, led to a series of new azaphenothiazine derivatives having 12(H)-quino[3,4-b][1,4] benzothiazine 4 structure. Reaction of compounds 4 in an alkaline milieu with alkylating agents occur as N-alkylation of the thiazine nitrogen and yields quinobenzothiazine derivatives 7. In vitro antiproliferative activity of compounds 4 and 7 was tested using two cancer cell lines (SNB-19 and C-32) and cisplatin as a reference. Most of the studied azaphenothiazine derivatives showed activity against both cell lines investigated (5.6–12.4 μg/ml concentration range tested). Compounds 4(b–e) containing a halogen atom or methyl group at the 9-position of the quinobenzothiazine ring show activity in the tested concentration range only against C-32 cell line. Compound 4f with methyl group in 11-position of quinobenzothiazine ring lacked activity against either cell line. The presence of additional aminoalkyl substituents at the thiazine nitrogen atom in compounds 7 increases their activity against both examined cell lines, when compared to compounds 4.

To improve the solubility, thermal properties and processability of polyxanthones, a new class of polyxanthones, poly(xanthone-imide)s (PXIs), with a high yield was prepared using a two-step chemical imidation of 2,7-diaminoxanthone with pyromellitic dianhydride (PMDA), 3,3',4,4'-benzophenone tetracarboxylic dianhydride (BTDA) and 2,2'-bis-(3,4-dicarboxyphenyl) hexafluoropropane dianhydride (6-FDA)). These PXIs were characterized with FT-IR and 1 H NMR spectroscopies. They presented a good solubility in aprotic polar solvents such as N,N-dimethyl acetamide (DMAc), N,N-dimethyl formamide (DMF), N-methyl pyrrolidone (NMP) and dimethyl sulfoxide (DMSO), and showed inherent viscosities in a range of 0.34-0.58 dL/g. These PXIs exhibited lower glass-transition temperatures than the original polyxanthones and a high thermal stability. The obtained results of the UV-vis absorption and photoluminescence indicated that the maximum absorption and fluorescence emission of PXIs were in the range of 300-304 nm and 432-510 nm, respectively. The antioxidant activity of PXIs was evaluated with a DPPH assay. The antioxidant values for PXIs were greater than for the parent xanthone (X). The polyimides were investigated for the extraction of environmentally noxious metal ions such as Cr (VI), Co (II), Ni (II), Cu (II), Pb (II) and Cd (II) from aqueous solutions.
