Synthesis and characterization of a novel pH-controllable composite hydrogel for anticancer drug delivery

Abstract: A novel pH-sensitive composite hydrogel (CH) namely 2-Acrylamido-2-methylpropane sulfonic acid (AMPS) grafted N-maleoylchitosan (MACTS) intercalated montmorillonite has been synthesized by in situ intercalation graft copolymerization of AMPS with MACTS intercalated MMT (MACTS/MMT) by using potassium persulfate (KPS) as a free radical initiator, in the presence of N,N-methylenebisacrylamide (MBA) as a crosslinking agent. A hydrophilic anticancer drug, 5-fluorouracil (5-FLU), has been chosen to investigate the l… Show more

“…In high-pH medium (7.4), the hydrophilic groups(-OH) on the composite were getting ionised and the electrostatic repulsion between these negative charges led to the high swelling of the composite; however, in acidic medium, the absence of swelling caused the collapse of AMCD-g-APTSB. Similar results were reported by Anirudhan and Sandeep [31].…”

“…The encapsulation efficiency of the composite depends on the amount of the monomers, cross-linking agent and also the pH of the medium. Acrylamido-2methylpropane sulfonic acid grafted N-maleoylchitosan intercalated montmorillonite showed encapsulation efficiency of 94% [31] whereas alginate/chitosan-coated mesoporous silica-based nanocarriers showed a value of 85.7% [36]. The AMCD-g-APSB (present study) showed higher encapsulation efficiency of 96.1% compared with other mesoporous silica materials.…”

“…90 Iron oxide (Fe 3 O 4 ) was covered with carboxymethyl chitosan (CMCS), 90 in which montmorillonite (MMT) was intercalated to enhance cellular uptake of the NPs. 90, 91 A lipophilic fluorescent dye, coumarin-6, was covalently intercalated to MMT to label the CMCS/MMT-covered MNPs for a cellular uptake study. 90, 92 Flow cytometry was used in quantifying the uptake of the coumarin-6 labeled CMCS/MMT-MNPs by HeLa cells.…”

“…125 The pHresponsiveness of hydrogels is usually attributed to their ionic groups. In particular, montmorillonite 126 and LAPONITE s127 are more or less pH-dependent due to the edge charges. The addition of these clay minerals can increase the ionization of polymers, following a chain expansion in response to pH.…”
