Synthesis and evaluation of radioiodinated terminal para-iodophenyl-substituted .alpha.- and .beta.-methyl-branched fatty acids

Abstract: Methods have been developed for the preparation of terminal p-idophenyl-substituted alpha- and beta-methyl-branched long-chain fatty acids. The syntheses and physical properties of 14-(p-iodophenyl)-2(RS)-methyltetradecanoic acid and 15-p-iodophenyl)-3(RS)-methylpentadecanoic acid are described. The radioiodinated agents are of interest as a result of the expected pronounced uptake and prolonged myocardial retention that may result from the inhibition of fatty acid metabolism. Tissue distribution studies in ra… Show more

“…Iodine 123-labeled BMIPP (I-123 BMIPP) is a modified fatty acid in which methyl branching has been introduced into the β -position to inhibit beta-oxidation in human myocardium, and has suitable characteristics for myocardial single-photon emission computed tomography (SPECT) because of its high uptake and longer retention in the myocardium [7,9].…”

“…Especially fluorine and (to a lesser extent) chlorine will exert their influence via such a mechanism. This effect becomes less pronounced for • Iodinated linoleic and linolenic acid gave results, indistinguishable from iodinated oleic acid' 31 . t LV: the uptake in the left ventricle (LV) is given.…”

“…Iodinated fatty acids are particularly useful for imaging because they are taken up by myocardium as normal, but are metabolised with relatively long retention in the myocardium [32]. Fatty acid is the normal substrate for energy production in the myocardium.…”
