Synthesis-Based Approach toward Direct Sandwich Immunoassay for Ciguatoxin CTX3C

Abstract: Ciguatoxins are the major causative toxins of ciguatera seafood poisoning. Limited availability of ciguatoxins has hampered the development of a reliable and specific immunoassay for detecting these toxins in contaminated fish. Monoclonal antibodies (mAbs) specific against both ends of ciguatoxin CTX3C were prepared by immunization of mice with protein conjugates of rationally designed synthetic haptens, 3 and 4, in place of the natural toxin. Haptenic groups that possess a surface area larger than 400 A(2) we… Show more

“…Preparation of 10C9 Fab-10C9 IgG was prepared as described previously (11). 10C9 Fab was obtained by papain digestion of 10C9 IgG and purified on a Protein A column (Fab preparation kit; Pierce) followed by cation exchange chromatography (Resource S column; GE Healthcare) and size exclusion chromatography (HiLoad TM 26/60 Superdex 200; GE Healthcare).…”

“…Other methods that use biological components have been developed for the detection of CTXs, such as RIA [167], ELISA [168][169][170][171], rapid enzyme immunoassay stick test [172,173] or fluorimetric assays based on changes in membrane potential [112]. CTXs can also be detected by LC-MS [174].…”

“…Orally, an amount as small as 70 ng of CTX 4 can induce intoxication symptoms in humans (16). To detect small amounts of ciguatoxins before ingestion of fish, highly sensitive antibodybased immunoassays have been necessitated; however, a reliable and easy-to-use assay kit is not currently available (17).…”
