Synthesis, Characterization, DNA Binding, and Photocleavage Activity of Oxorhenium (V) Complexes with α‐Diimine and Quinoxaline Ligands

Abstract: The complex [ReOCl3pq] (1) (where pq = 2-(2′pyridyl)quinoxaline) has been synthesized and fully characterized by UV-Vis, FTIR, 1 and 2D NMR, and cyclic voltammetry (CV). The DNA-binding properties of the complex 1 as well as of the compounds [ReOCl3bpy] (2), [ReOCl3phen] (3), and pq (4) were investigated by UV-spectrophotometric (melting curves), CV (cyclic voltammetry), and viscosity measurements. Experimental data suggest that complex 1 intercalates into the DNA base pairs. Upon irradiation, complex 1 was fo… Show more

“…ReOCl 3 (PPh 3 ) 2 was prepared as previously reported. 78 The synthesis of 1 has been reported previously, 79 but the procedure reported here is novel. Analytical grade acetone and dichloromethane were used as solvents.…”

“…The corresponding bidentate ligand has been chosen owing to the remarkable biological properties reported [125, 126], while, on the other hand, investigation of the biological activity of inert metal complexes, such as rhodium(III) compounds, shows gradually increasing interest [127130]. These reasons led us to design the target molecule Rh-1 (Figure 4) with octahedral coordination geometry and evaluate in vitro its inhibitory effect against PAF-induced platelet aggregation in washed rabbit platelets (WRPs).…”

“…In 2009, we have successfully reported a preliminary work [ 124 ] on the anti-PAF activity of a Rh(III) coordination compound, namely, cis-[Rh(L) 2 Cl 2 ]Cl ( Rh-1 ), where L stands for the bidentate ligand 2,2′-pyridylquinoxaline with an IC 50 value of 250 nM ( Figure 4 ). The corresponding bidentate ligand has been chosen owing to the remarkable biological properties reported [ 125 , 126 ], while, on the other hand, investigation of the biological activity of inert metal complexes, such as rhodium(III) compounds, shows gradually increasing interest [ 127 130 ]. These reasons led us to design the target molecule Rh-1 ( Figure 4 ) with octahedral coordination geometry and evaluate in vitro its inhibitory effect against PAF-induced platelet aggregation in washed rabbit platelets (WRPs).…”

“…When coordinated to metal ions, the resulting complexes have interesting bioinorganic applications. For example, [ReOCl 3 (2, 2′-pq)] can efficiently cleave plasmid pBR 322DNA upon irradiation by generating singlet oxygen and hydroxyl radicals [18]. Furthermore, incorporation of 2, 2′pq in Pt-diamine complexes [19] and study against murine leukemia results in IC 50 values in the range of ∼40 μM.…”
