“…In addition, monolayer 2-dimensional culture induces loss of the differentiated phenotype, as characterized by the upregulation of col1 and the downregulation of col2 [Benya and Shaffer, 1982;Zwicky and Baici, 2000]. Some previously created immortalized cell lines have been incapable of producing col2 [Takigawa et al, 1989;Block et al, 1991;Thenet et al, 1992], and therefore both the phenotype and morphology of A13/BACii were investigated and compared to freshly isolated bovine chondrocytes and the col2-producing cell line C-20/A4 [Goldring et al, 1994]. As 3-dimensional encapsulation into supports composed of col2, agarose or alginate enables the recapture of phenotype [Beekman et al, 1997;Nehrer et al, 1997;Benz et al, 2002;Miot et al, 2005], we investigated the changes in both the morphology and 'expression battery' of A13/BACii in response to HD low-melt agarose 3-dimensional culture as previously described [Schulze-Tanzil et al, 2002].…”

“…Two lines of evidence suggest that regulation of GAG chain attachment may be important during tissue development. It has been reported that attachment of GAG chains changes during tissue growth, and the types and numbers of GAG chains vary during different growth stages (34). Furthermore, the GAG chains of versican have been reported to be involved in reducing cell adhesion (35).…”

“…It is, therefore, important to establish a model of CS with a chondrosarcomatous phenotype for clinical investigation. Until now, there have been some reported cell lines of CS; however, those with chondrocytic phenotypes were limited [4,10,11,24,45]. In this study, new cell lines with chondrocytic phenotypes were established from a pelvic chondrosarcoma, and the morphological and molecular analyses were performed.…”
