BACKGROUND Cross‐linking enzyme aggregates (CLEAs) seem to be a promising immobilization method due to mild processing conditions and short preparation time. However, the CLEAs prepared in aqueous solution exhibit amorphous large clusters that cause significant mass‐transfer limitations, leading to low catalytic efficiency. In this work, novel spherical CLEAs of lipase from bovine pancreas (spherical CLEAs) were prepared within droplets dispersed in a water‐in‐hydrophobic ionic liquid microemulsion rather than a conventional aqueous solution. The properties of the spherical CLEAs were determined. RESULTS The resultant CLEAs exhibited spherical structure with good monodispersity instead of the amorphous clusters of conventional CLEAs, and showed higher activity recovery (84.6%) than that of conventional CLEAs (52.8%). The high activity recovery of spherical CLEAs could be attributed to both interfacial activation of the microemulsion system and the well‐defined structure of the spherical CLEAs, which not only activated lipases in an active form, but also decreased mass‐transfer limitations. Moreover, the thermostability, storage stability, tolerance against hydrophilic solvents, and mechanical stability of the spherical CLEAs were enhanced significantly. The spherical CLEAs evidenced excellent reusability in comparison with conventional CLEAs. CONCLUSIONS The novel strategy in this study might open an attractive way towards effectively improving the catalytic properties of traditional CLEAs. © 2016 Society of Chemical Industry

Bio-based fatty acids (FAs) produced through hydrolysis of natural oils and fats are promising chemical feedstocks for increasing the economic value of renewable raw materials. In this work, lecithin, gelatin, PEG, and MgCl2 were employed as the co-immobilized material of crude lipase Mucor miehei immobilization on the polyurethane foam (PUF) matrix for hydrolysis of coconut oil to Free Fatty Acid (FFA). The unconventional immobilized technique was used through cross-linking and covalent bond. Single factor analysis and response surface method were utilized to determine the optimum conditions of the hydrolysis reaction. After optimization, co-immobilized lipase was examined for storage stability at a temperature of 4°C and reusability performance. The optimum conditions for coconut oil hydrolysis were obtained on the co-immobilized-PUF ratio, water-oil ratio, and reaction time of 20.17 w/w, 4.45 w/w, and 20 h, respectively. Under these conditions, the acid value as lauric acid enhanced 573% to 3.21 mg KOH/g oil. Storage stability attained through remaining activity on free lipase, PUF-lipase, PUF-co-immobilized-lipase were 9.89%, 42.3%, and 91.88%, respectively. In this study, the application of PUF-co-immobilized lipase in hydrolysis reactions can be reused up to 5 times. Characteristics of the addition of co-immobilized lipase have been analyzed using Fourier Transform Infra Red (FTIR) and Scanning Electron Microscope (SEM), showing the presence of functional groups binding and the changes in the surface matrix structure.

Biotransformation of Plant-Derived Phenolic Acids

Phenolic acids are abundant biomass feedstock that can be derived from the processing of lignin or other byproducts from agro-industrial waste. Although phenolic acids such as p-hydroxybenzoic acid, p-coumaric acid, caffeic acid, vanillic acid, cinnamic acid, gallic acid, syringic acid, and ferulic acid can be used directly in various applications, their value can be significantly increased when they are further modified to high value-added compounds. This review summarizes and discusses the new advances in cell-free and whole-cell biocatalysis technologies for reactions important for conversion of phenolic acids including esterification, decarboxylation, amination, halogenation, hydroxylation, and ring-breakage reactions. The products of these reactions are useful for the pharmaceutical, cosmetic, food, fragrance, and polymer industries. Production of phenolic acids is sustainable, and these processes for their biotransformation are clean technologies that do not produce toxic waste and use less energy than conventional physical and chemical methods. Thus, biotransformation of phenolic acids provides an economically viable and sustainable means for producing useful materials for society.
