Synthesis of Dextran–Phenoxodiol and Evaluation of Its Physical Stability and Biological Activity

Abstract: Phenoxodiol, an isoflavene anti-tumor agent, was conjugated on the polysaccharide dextran using immobilized laccase as biocatalyst. The success of the enzymatic conjugation was determined by UV-vis spectrophotometry and its functionalization degree was assessed by 1 H NMR and was found to be 3.25 mg phenoxodiol/g of conjugate. An accelerated stability test showed that the resultant conjugate was nine times more stable than the free phenoxodiol when tested for its residual anti-oxidant ac… Show more

“…As the polymer slowly degrades under acidic pH (Javid et al, 2013), the therapeutic loads of the antitumoral agents (DOX and PTX, in our study) are maintained for a longer time period, making the therapy very convenient for patient use (Duncan, 2006; Ketabat et al, 2019; Singh et al, 2019). The OVCAR‐3 cells grew aggressively inside the SCID mice body but the antitumoral effect of DOX–AP–SPIO and DOX–CS–SPIO was identical on both of the groups (A2780‐Balb/c and OVCAR‐3‐Balb/c) (Hine, Seluanov, & Gorbunova, 2012; Yee et al, 2019).…”

“…The anti-cancer activity was also tested in HMEC-1 (human microvascular endothelial cells) and non-malignant human lung fibroblast MRC-5 cells. This conjugate was more stable, effective and less toxic than the free drug [ 127 ].…”

“…Dextran has decent water solubility and flexibility for functional modifications. Dextran can stabilize the DDSs or drug conjugates and enhance the bioavailability of a drug after administration without significant toxicity [28]. Therefore, it and its derivatives are suitable for developing as drug carriers such as micelles and hydrogels.…”
