Synthesis of Novel Hantzsch Dihydropyridine Derivatives

Abstract: A one-pot synthesis of novel Hantzsch dihydropyridine derivatives via a three-component cyclocondensation reaction of p-nitro acetoacetanilide, aromatic aldehyde and aqueous ammonia solution is presented. The protocol avoids the use of expensive catalyst, toxic solvent and chromatographic separation.

“…Catalysis with PPA allowed to increase the yields of reaction products 9a,b to 18 and 27%, respectively, but led to problems with product isolation from the reaction mixture. By analogy to published procedures, 15,16 where the active component (PPA) was immobilized on silicon dioxide surface, we obtained a solid acidic catalyst by using nanosized ТiO 2 -SiO 2 as a support for PPA. Indeed, the PPA-modified nanosized ТiO 2 -SiO 2 oxide enabled a more selective interaction of podands 5a,b with urea (7) and benzaldehyde (8), producing the target dihydropyrimidine derivatives 9a,b in 41 and 52% yields, respectively.…”
