Synthesis of novel oligomeric anionic alkyl glycosides using laccase/TEMPO oxidation and cyclodextrin glucanotransferase (CGTase)‐catalyzed transglycosylation

Abstract: Modification of alkyl glycosides, to alter their properties and widen the scope of potential applications, is of considerable interest. Here, we report the synthesis of new anionic alkyl glycosides with long carbohydrate chains, using two different approaches: laccase/2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) oxidation of a long-carbohydrate-chain alkyl glycoside and cyclodextrin glucanotransferase (CGTase)-catalyzed elongation of anionic alkyl glycosides. The laccase/TEMPO oxidation of dodecyl β-D-maltooct… Show more

“…It is of great interest to note that both enzymes efficiently catalyze the transglucosylation of baicalin. This confirms the recent findings of Ngo et al [17], namely, that glucuronide can be glucosylated by the CGTase from Thermoanaerobacter sp. In addition to the high yield of transglusylation afforded by Toruzyme ® , the enzyme was more tolerant to pH variations than CGTase Amano in the range from 4 to 6 (Supplementary Data S1).…”

“…Ngo et al recently demonstrated that CGTase from Thermoanaeroacter sp. is able to transfer a glucosyl unit from α-cyclodextrin to octyl β-D-glucuronic acid on the C4 position of glucuronide [17], suggesting that baicalin could be an acceptor of this enzyme.…”

“…Some examples with main specificities for α-glucosyl groups, include cyclomaltodextrin glucanotransferases (CGTases in family GH13) [5,6]. CGTases can also be used for the synthesis of alkyl glycosides [7,8].…”
