Synthesis of triacylglycerol containing conjugated linoleic acid by esterification using two blended lipases

Abstract: We have developed an efficient esterification for the synthesis of triacylglycerol (TAG) containing conjugated linoleic acids (CLA) using a blend of two powdered lipases. Two pairs of blended lipases promoted the esterification. Rhizomucor miehei lipase plus Alcaligenes sp. lipase and Penicillium camembertii MAG and DAG lipase plus Alcaligenes sp. lipase were used. At the optimal ratio of two lipases, the content of TAG containing CLA (TAG-CLA) in all glycerols reached 82-83% after 47 h using 1 wt% of lipases.… Show more

“…An interesting idea for improving the content of CLA in structured triacylglycerols to about 83% has been presented by Hirose et al [129], in which two lipases, one with a high degree of sn-1,3 selectivity (i.e. Rhizomucor miehei or Penicillium camembertii lipase) and a second lipase with a lower selectivity towards the sn-1,3-position (lipase from Alcaligenes sp.…”

“…This could be because higher temperatures caused adverse effects on enzyme activity through changing enzyme conformation. One study applied 40°C as the catalysis temperature for Palatase in a solvent-free system under high operation pressure (Hirose, Yamauchi-Sato, Arai, & Negishi, 2006). The different observations in that study may be due to these different reaction systems and conditions.…”

“…This is followed by two references for the preparation of STAG containing the esters of CLA (Entries 28 and 29; Arcos and Hill, 2000;Hirose et al, 2006). The various isomers of CLA have been reported to have numerous beneficial effects on human health and thus it is likely that research into CLA-containing lipids will continue.…”
