Mononuclear complexes of Co(II), Ni(II), Cu(II), Zr(IV), Pd(II), and Cd(II) with metformin hydrochloride (MF) and 1,4-diacetylbenzene (DAB) have been synthesized, and their structures have been elucidated on the basis of FT-IR, 1 H NMR, and electronic spectra, molar conductance, magnetic susceptibility measurements, thermal, and elemental analysis. Molar conductance measurements indicate electrolytic nature of the complexes. According to FT-IR spectra, MF reacts with metal ions as a bidentate ligand through its two imino groups, and DAB reacts as a monodentate ligand through one of two equivalent keto groups. The complexes have octahedral structure, except Pd(II) complex which is square planar. TGA and DTG analyses support the suggested stereochemistry and subsequently the proposed mechanism of thermal decomposition. Coats-Redfern and Horowitz-Metzger equations have been used for calculating kinetic and thermodynamic parameters of the main decomposition step. Molecular modeling calculations confi rm the structure of the complexes. The ligands and their complexes in vitro antimicrobial activity tests against some bacterial and fungal strains indicate higher activity of the complexes than that of MF and DAB.

Excessive activities of cysteinyl cathepsins (CysCts) contribute to the progress of many diseases; however, therapeutic inhibition has been problematic. Zn 2 + is a natural inhibitor of proteases with CysHis dyads or CysHis(Xaa) triads. Biguanide forms bidentate metal complexes through the two imino nitrogens. Here, it is discussed that phenformin (phenylethyl biguanide) is a model for recruitment of endogenous Zn 2 + to inhibit CysHis/CysHis(X) peptidolysis. Phenformin is a Zn 2 + -interactive, anti-proteolytic agent in bioassay of living tissue. Benzoyl- l -arginine amide (BAA) is a classical substrate of papain-like proteases; the amide bond is scissile. In this review, the structures of BAA and the phenformin-Zn 2 + complex were compared in silico. Their chemistry and dimensions are discussed in light of the active sites of papain-like proteases. The phenyl moieties of both structures bind to the “S2” substrate-binding site that is typical of many proteases. When the phenyl moiety of BAA binds to S2, then the scissile amide bond is directed to the position of the thiolate-imidazolium ion pair, and is then hydrolyzed. However, when the phenyl moiety of phenformin binds to S2, then the coordinated Zn 2 + is directed to the identical position; and catalysis is inhibited. Phenformin stabilizes a “Zn 2 + sandwich” between the drug and protease active site. Hundreds of biguanide derivatives have been synthesized at the 1 and 5 nitrogen positions; many more are conceivable. Various substituent moieties can register with various arrays of substrate-binding sites so as to align coordinated Zn 2 + with catalytic partners of diverse proteases. Biguanide is identified here as a modifiable pharmacophore for synthesis of therapeutic CysCt inhibitors with a wide range of potencies and specificities. Graphical abstract Phenformin-Zn 2+ Complex

Complexes of Nd(III) and Sm(III) of a Schiff base ligand derived from the condensation of o-phenylenediamie and 2-hydroxybenzaldehyde were synthesised and characterised by various analytical and spectroscopic techniques. The analytical values revealed a metal to ligand ratio of 1:2 for the Nd(III) complex and a 1:1(ligand: metal) for Sm(III) complex. The complexes were characterised by elemental analysis, molar conductance, UV – Visible and infrared spectral studies. Plausible structures were proposed for Nd(BSOPD)2 and SmBSOPD based on the results obtained. In-vitro antimicrobial activities of both the Schiff base and the synthesised metal complexes were evaluated against six (6) human pathogens. The complexes showed significant activity compared to the free ligand. These complexes may be employed as active ingredients in the development of antibacterial and antifungal agents.
