Synthetic Circular miR-21 Sponge as Tool for Lung Cancer Treatment

Abstract: Lung cancer is the most common cancer in the world and several miRNAs are associated with it. MiRNA sponges are presented as tools to inhibit miRNAs. We designed a system to capture miRNAs based on circular RNAs (circRNA). To demonstrate its usefulness, we chose miR-21, which is upregulated and implicated in lung cancer. We constructed a miR-21 sponge and inserted it into a vector that facilitates circular RNA production (Circ-21) to study its effect on growth, colony formation, and migration in lung cancer ce… Show more

“… 35 Rama et al. 36 established a vector-based protocol using ALU sequences to generate a miR decoy with seven binding sites for miR-21 to suppress tumorigenesis in lung cancer cells. Nevertheless, back-splice-imitating constructs are limited in their ability to be delivered using lentiviral particles.…”

“…To determine expression, RNA was extracted from both cell lines with the miRCURY LNA miRNA PCR Starter Kit (Qiagen). To carry out the quantitative real-time polymerase chain reaction (qRT-PCR), we followed the protocol described by Rama et al [ 31 ] using miR-103a and miR-191 using miR-103a and miR-191 as housekeeping.…”

“…We used the pcDNA3.1(+) CircRNA mini vector (Addgne), in which a sponge against miR-21 (Circ-21) had been subcloned, and on the other hand, the enhanced green fluorescent protein ( EGFP ) gen (Circ-EGFP), which was used as the reporter. Both of them were designed and constructed by Rama et al [ 31 ]…”
