??-Synuclein Protein Levels Are Increased in Alcoholic Patients and Are Linked to Craving

Abstract: To our knowledge, this is the first study evaluating alpha-synuclein protein expression in alcoholics. The current study provides further evidence of altered alpha-synuclein levels in patients with alcoholism and their linkage to alcohol craving. Because alpha synuclein is involved in the modulation of dopaminergic neurotransmission, these results deliver further pathophysiological explanations of craving mechanisms.

“…␣-Synuclein also regulates DAT (Wersinger et al, 2003), and the increased expression of ␣-synuclein mRNA in our heroin subjects is comparable with other substance abuse human studies showing elevated ␣-synuclein mRNA and protein levels in cocaine abusers (Mash et al, 2003;Qin et al, 2005) and alcoholics (Bonsch et al, 2004(Bonsch et al, , 2005. Increased midbrain ␣-synuclein is characteristic of neurodegeneration of DA pigmented cells (elAgnaf and Irvine, 2002) within the substantia nigra.…”

“…21 a-Synuclein inhibits the enzyme tyrosine hydroxylase, thereby downregulating the synthesis of dopamine (DA). 22,23 a-Synuclein is implicated in neurodegenerative disorders 24 such as Parkinson's disease, [25][26][27][28] and high levels of a-synuclein mRNA have been associated with alcoholism in humans 29,30 and alcohol preference in rats. 31 Since anxiety is a common feature in Parkinson's patients, 32 and also in alcoholics, 33,34 a-synuclein may also be important in anxiety.…”

“…Later work demonstrated a correlation between the levels of alpha synuclein in serum with the intensity of alcohol craving in addicted patients during withdrawal (Bonsch et al, 2005a;Bonsch et al, 2004). Monkeys that chronically selfadminstered ethanol for 18 months had 3-fold higher peripheral blood levels of alpha synuclein mRNA, suggesting that peripheral alpha synuclein upregulates with chronic exposure to alcohol (Walker and Grant, 2006).…”

“…This region has been associated with drug abuse through a genome-wide SNP genome scan (Uhl et al, 2001). Genomic variation in SNCA has been associated with susceptibility to alcohol (Bonsch et al, 2005b), methamphetamine psychosis/dependence (Kobayashi et al, 2004) and alcohol craving (Bonsch et al, 2005a;Bonsch et al, 2005b;Foroud et al, 2000;Foroud et al, 2007). Using a family based test of association in multiplex alcoholic pedigrees, analyses in the 3′ end of SNCA identified a haplotype overestimated in European-American individuals who crave alcohol and a different haplotype, which was preferentially transmitted to noncravers (Foroud et al 2007).…”
