Systematic Analyses of the Transcriptome, Translatome, and Proteome Provide a Global View and Potential Strategy for the C-HPP

Abstract: To estimate the potential of the state-of-the-art proteomics technologies on full coverage of the encoding gene products, the Chinese Human Chromosome Proteome Consortium (CCPC) applied a multiomics strategy to systematically analyze the transciptome, translatome, and proteome of the same cultured hepatoma cells with varied metastatic potential qualitatively and quantitatively. The results provide a global view of gene expression profiles. The 9064 identified high confident proteins covered 50.2% of all gene p… Show more

“…Favorable to this rationale, we previously defined a specific preference of translation initiation on genes that regulate cellular movement and cytoskeleton, with over 4-fold greater translation ratios in cancer cells than those in normal epithelial cells [5153]. Furthermore, active secretion behavior has been recognized as an important feature of CRC cells [54, 55].…”

“…Increasing numbers of investigators are now incorporating RNA-Seq information with proteomic data to gain a more complete understanding of cellular systems and improve genome annotation (19)(20)(21). The comparative analysis of RNA and protein levels can be used as a validation tool to generate a protein atlas with higher reliability.…”

“…These low-abundance proteins are likely to exhibit their transcriptional expression at both tissue and subcellular specific manner (PE2 proteins) and may be involved in gene regulation in poorly understood regions of the genome [53]. Because of the large dynamic range of proteins in biological samples, low-abundance isoforms and proteoforms that may be associated with disease, can be extremely difficult to measure [54].…”
