Systemic antibiotic therapy does not significantly improve outcome in a rat model of implant-associated osteomyelitis induced by Methicillin susceptible Staphylococcus aureus

Abstract: Systemic perioperative and implant-associated application of antibiotics were both comparably effective to treat implant-associated infections whereas the combined antibiotic therapy further reduced systemic signs of infection time dependent.

“…213215 Gentamicin is the most studied antibiotic in coatings because of its application in the clinic for treating methicillin-sensitive S. aureus (MSSA) bone-related infections. 203,204,206208,210,211,213,214,216218 Vancomycin, commonly used to treat MRSA infections, has also been incorporated into implant coatings. 212,215,219 Additional antibiotics such as fosfomycin, 204 doxycycline, 209 minocycline, 20 rifampin, 20 colistin, daptomycin, and cefoxitin have also been incorporated into implant coatings.…”

“…Haptoglobin has been used to track infection and success of treatment in 5‐month‐old male Sprague Dawley rats receiving femoral injections of methicillin‐susceptible S . aureus (MSSA) into the intermedullary cavity 48 . Rats receiving a gentamicin‐coated K‐wire implant with intraperitoneal (IP) gentamicin sulfate injection (combination group) showed decreased levels of haptoglobin on Days 7, 28, and 42 compared to uncoated implant alone (control) 48,49 .…”

“…The increasing number of joint replacement revision surgery for infectious disease is challenging (Frommelt 2018 ; Li et al 2013 ). Local application of antibiotics within impacted bone graft appears useful in case of prevention or treatment of infection (Coraca-Huber et al 2016 ; Fölsch et al 2015 , 2016a ; Lewis et al 2012 ). Suitable carrier substances provide high local antibiotic concentrations (Frommelt 2018 ) and should improve mechanic properties of the impacted bone graft.…”
