Systemic sclerosis: Current concepts in pathogenesis and therapeutic aspects of dermatological manifestations

Abstract: Systemic sclerosis (SSc) is a chronic, multisystem connective tissue disease with protean clinical manifestations. Recent advances in understanding the pathogenic mechanisms have led to development of target-oriented and vasomodulatory drugs which play a pivotal role in treating various dermatological manifestations. An exhaustive literature search was done using Medline, Embase, and Cochrane library to review the recent concepts regarding pathogenesis and evidence-based treatment of salient dermatological man… Show more

“…In addition to these, HBOT has been shown to promote angiogenesis that is demonstrated to be strongly disturbed in SSc patients and stimulate tissue growth by regulating various growth factors and metalloproteinases . Also, HBOT is known to help antimicrobial activity and regulate inflammatory cytokines, fibroblast proliferation, and collagen formation .…”

“…Steroids inhibit collagenase activity. 3 Corticosteroids also prevent new blood vessel formation by inhibiting expression of the vascular endothelial growth factor gene in human vascular smooth muscle cells. 17 Scleroderma, or SS, is characterised by increased deposition of hyaluronic acid (HA) and collagen.…”

“…The key cellular moderator of fibrosis is collagen-producing myofibroblasts. 3 Usually, the diagnosis of systemic scleroderma is clinical. Effective treatment for scleroderma still remains inconclusive.…”
