Systems analysis of subjects acutely infected with the Chikungunya virus

Abstract: The largest ever recorded epidemic of the Chikungunya virus (CHIKV) broke out in 2004 and affected four continents. Acute symptomatic infections are typically associated with the onset of fever and often debilitating polyarthralgia/polyarthritis. In this study, a systems biology approach was adopted to analyze the blood transcriptomes of adults acutely infected with the CHIKV. Gene signatures that were associated with viral RNA levels and the onset of symptoms were identified. Among these genes, the putative r… Show more

“…No mouse models have so far been developed that measure polyarthralgia. The adult wild-type CHIKV mouse model recapitulates many aspects of human disease [ 42 , 57 ], with recent RNA-Seq comparisons further illustrating that this mouse model shares many inflammatory processes with those seen in humans [ 105 , 106 ]. However, such comparisons are not yet available for ONNV, MAYV or RRV.…”

“…The mechanisms whereby the high fiber diet would mediate the neutrophil-promoting changes during CHIKV arthritis are likely to be complex given the interaction of various SCFAs with multiple cell types (10, 18, 24, 66, 113), the infection of different cell types by CHIKV, and the complex interplay of anti-viral and inflammatory responses seen post infection (36, 41, 42, 51). Nevertheless, a likely key contributing mechanism highlighted herein is the down-modulation in the rM gene signature seen in feet during peak CHIKV arthritis in mice fed a high fiber diet (compared with peak CHIKV arthritis in mice fed a no fiber diet) (Figure 5).…”

“…CD4 T cells are important for driving CHIKV arthritis (36), with Tregs associated with disease amelioration (49, 50). Macrophages/monocytes also play an important role in the arthritic immunopathology (36), with the pro-inflammatory response to CHIKV infection in peripheral blood shown to be monocyte centric (41, 51). However, macrophages are also required for resolution of inflammation, both generally (5254) and specifically for CHIKV arthritic inflammation (45).…”
