Take Advantage of Glutamine Anaplerosis, the Kernel of the Metabolic Rewiring in Malignant Gliomas

Abstract: Glutamine is a non-essential amino acid that plays a key role in the metabolism of proliferating cells including neoplastic cells. In the central nervous system (CNS), glutamine metabolism is particularly relevant, because the glutamine-glutamate cycle is a way of controlling the production of glutamate-derived neurotransmitters by tightly regulating the bioavailability of the amino acids in a neuron-astrocyte metabolic symbiosis-dependent manner. Glutamine-related metabolic adjustments have been reported in s… Show more

“…Thus, considerable and continuous efforts are being directed toward exploiting specific dependencies to identify novel molecular targets and design new effective therapeutic strategies that help improve the patient’s clinical care with GBM ( 8 , 35 , 122 ). Indeed, the paradigm of focusing on cancer metabolism was freshly required beyond the dramatic flexibleness of tumor metabolic cascades reported in cancer cells challenged with multiplex inhibitors, including glycolysis and glutaminase inhibitors ( 123 , 124 ).…”

“…Cells having different metabolic phenotypes, with or without TRAP1, were forced to rely on either glucose, pyruvate, or glutamine as the sole carbon source. Surprisingly, consistent among all the cell types considered, TRAP1-deficient cells were unable to support OxPhos with either glucose or pyruvate, instead relying on glutamine, which served as an anaplerotic molecule [ 95 ] to support the TCA cycle and OxPhos upon conversion to α-ketoglutarate in mitochondria. Confusingly, all these metabolic behaviors are pro-neoplastic [ 85 , 96 , 97 , 98 ].…”

“…A continuous supply of precursors, sustained by anaplerotic pathways, “refills” the cycle with intermediates to replace those that have been already used for biosynthesis. Glutaminolysis, which produces α-ketoglutarate from glutamine, and pyruvate carboxylation, which produces oxaloacetate from glucose/pyruvate are major contributors to anaplerotic fluxes in cancer cells ( Bott et al, 2019 ; Martins et al, 2020 ; Kiesel et al, 2021 ). Oxidation of the branched-chain amino acids (BCAAs) isoleucine and valine also provides an anaplerotic flux in some tumors.…”
