Target delivery of MYCN siRNA by folate-nanoliposomes delivery system in a metastatic neuroblastoma model

Abstract: BackgroundFolate-nanoliposomes delivery system has emerged recently as a specific and safety delivery method and gradually used as the carrier of a variety kinds of drugs including compounds, plasmids and siRNAs.MethodsIn this study, we established a bone marrow and bone metastasis xenograft mouse model by injecting the LA-N-5 cell into the bone marrow cavity. Fluorescence microscopy, TUNEL Assay, Quantitative RT-PCR and western blot were conducted to analysis the distribution of folate-nanoliposomes entrapped… Show more

“…In our study, Oct4‐positive cells were found more frequently in tumors with unfavorable group compared with favorable group and the presence of Oct4‐positive cells was more frequent in higher stages in comparison with lower stages (23 cases vs 4, respectively, although no significant relationship was found). Oct4 is one of the factors, which repress the differentiation process in the blastocytes of preimplanted embryo and is required to activate genes involving in early development . Newton et al.…”

“…A modified nanoprecipitation method was used to entrap DM1 into the FA-PLA-TPGS NPs [ 23 , 24 ]. Briefly, a pre-weighed amount of DM1 drug powder and 100 mg of FA-PLA-TPGS copolymer were dissolved in 8 mL of acetone by vortexing and sonication.…”

“…Results suggest that PELP1-siRNA-DOPC nanoliposomes can be used as a potential therapeutic modality for the prevention or treatment of ovarian cancer metastasis [105]. Several other siRNA-liposomal formulations targeting key genes involved in cancer growth, proliferation, drug resistance, and metastasis are under investigation [106][107][108][109][110].…”
