Targeting Phosphoinositide 3-Kinase γ in Airway Smooth Muscle Cells to Suppress Interleukin-13-Induced Mouse Airway Hyperresponsiveness

Abstract: We recently reported that phosphoinositide 3-kinase ␥ (PI3K␥) directly regulates airway smooth muscle (ASM) contraction by modulating Ca 2ϩ oscillations. Because ASM contraction plays a critical role in airway hyperresponsiveness (AHR) of asthma, the aim of the present study was to determine whether targeting PI3K␥ in ASM cells could suppress AHR in vitro and in vivo. Intranasal administration into mice of interleukin-13 (IL-13; 10 g per mouse), a key pathophysiologic cytokine in asthma, induced AHR after 48 h… Show more

“…Indeed, reduction of AHR was observed only when the NOP receptor was activated in vivo by administering it to mice at days 0 and 7, 30 min prior to the allergen administration. Moreover, in our model, in agreement with several studies (15,19,31), IL-13 was able to induce AHR. UFP-112 treatment 30 min before IL-13 administration was unable to modify IL-13-induced AHR, further confirming the role of the NOP receptor agonist in the early phases of the sensitization process.…”

“…Some authors indicate that eosinophil infiltration has an important role in the development of AHR [30], while others demonstrate a close association between AHR and T helper lymphocytes [29]. Moreover, hyperreactivity is caused by a direct effect of IL-13 on airway smooth muscle [31]. In contrast to the study from Wood and colleagues, we report that FO intake improved resistance and elastance [15].…”

“…Cells were seeded on 6-well plates. The following day, adherent MDA-MB-231 cells were re-transfected with the same siRNA using Lipofectamine 2000 as previously described [32]. Two days later, cells were harvested for Western blot analysis of PI3Kα, PI3Kβ or PI3Kγ expression or migration and invasion assays.…”
