Targeting single-walled carbon nanotubes for the treatment of breast cancer using photothermal therapy

Abstract: This paper focuses on the targeting of single-walled carbon nanotubes (SWNTs) for the treatment of breast cancer with minimal side effects using photothermal therapy. The human protein annexin V (AV) binds specifically to anionic phospholipids expressed externally on the surface of tumour cells and endothelial cells that line the tumour vasculature. A 2 h incubation of the SWNT-AV conjugate with proliferating endothelial cells followed by washing and near-infrared (NIR) irradiation at a wavelength of 980 nm wa… Show more

“…A5-mβG binding matched previously reported K d values of A5 alone binding to cell surface phosphatidylserine, ranging from 0.1 to 2 nM (Schutters and Reutelingsperger, 2010;Neves et al, 2013). As expected, A1-mβG exhibited higher K d values than A5-mβG.…”

“…However, no uptake studies were performed on these cells; it would be of particular interest if the same preferential uptake observed by us was also observed with graphene nanoparticles. The efficacy of CNTs and NIR was reported for different types of cancer (6, 2123). However, it is still unclear if this strategy can be used for glioblastoma.…”

“…Neves et al functionalized SWNTs with annexin V (AV) to target the tumor vasculature of breast cancer cells. 61 In vitro studies with HAAE-1 human endothelial cells showed selective killing compared to cells treated with the laser alone. Since cells were washed thoroughly after treatment with SWNT-AV conjugates, it was clear that cell death resulted from binding of SWNT-AV to the cells.…”
