Targeting the Oncogenic Transcriptional Regulator MYB in Adenoid Cystic Carcinoma by Inhibition of IGF1R/AKT Signaling

Abstract: MYB-NFIB is an oncogenic driver and a key therapeutic target in ACC that is regulated by AKT-dependent IGF1R signaling. Our studies uncover a new strategy to target an oncogenic transcriptional master regulator and provide new important insights into the biology and treatment of ACC.

“…3a). To confirm that our models are representative of MYBinduced gene expression in ACC, we investigated the association between gene sets derived from MYB/MYB-NFIB overexpressing MCF10A cells compared with a previously published gene expression data set 16 of MYBpositive ACC patient samples ( Supplementary Fig. 2).…”

“…5). To investigate whether ATR is a downstream target of MYB-NFIB in ACC, we analyzed global gene expression data sets with siRNA knockdown of MYB-NFIB in cultured ACC cells 16 . Treatment with MYB siRNA caused a significant decrease in the expression of ATR, suggesting that MYB-NFIB regulates ATR expression in ACC cells (Fig.…”

“…Ten retrospective studies including 658 patients with ACC fulfilled the inclusion criteria and were included in this meta-analysis ( Figure 1). 12,15,17,18,[21][22][23][24][25][26] Three studies without outcome events were excluded, 16,27,28 and one study that investigated the impact of MYB mRNA on prognosis was also excluded. 29 The characteristics of the 10 eligible studies are provided in Table 1.…”
