Tautomeric transformations of piroxicam in solution: a combined experimental and theoretical study

Abstract: Piroxicam tautomerism was studied in solution. Enol-amide tautomer is presented mainly as a sandwich type dimer in ethanol and DMSO. The addition of water leads to gradual shift of the equilibrium towards the zwitterionic tautomer.

“…It shows very good results in predicting the position of tautomeric equilibria for compounds with intramolecular hydrogen bonds. 14,[24][25][26][27] For comparison some additional functionals as implemented in Gaussian 09 D.01 (B3LYP 28 , MN12SX 29 , BHandH 30 ) were used as well. Second and higher order Møller-Plesset perturbation theory (MP2-4) 31 and coupled-cluster theory with single, double, and perturbative triple excitations (CCSD(T)) 32 were used in addition for calculations of the ground state.…”

“…Benzosultams, a subclass of bicyclic sulfonamides, are found in numerous biologically active compounds, pharmaceuticals, as well as agricultural agents [ 3 , 4 , 5 , 6 , 7 ]. They exhibit a wide spectrum of promising bioactivities, such as antibacterial, antidiabetic, anticancer, and vitro HPPD (4-hydroxyphenylpyruvate dioxygenase) inhibitory activities, which have been widely utilized as inhibitors and drugs [ 8 , 9 , 10 , 11 , 12 ]. Representative examples have been outlined in Scheme 1 .…”
