Taxol binds to cellular microtubules.

Abstract: ABSlRACT Taxol is a low molecular weight plant derivative which enhances microtubule assembly in vitro and has the unique ability to promote the formation of discrete microtubule bundles in cells. Tritium-labeled taxol binds directly to microtubules in vitro with a stoichiometry approaching one (Parness, J., and S. 8. Horwitz, 1981, J. Cell Biol. 91:479-487). We now report studies in cells on the binding of [aH]taxol and the formation of microtubule bundles.[aH]Taxol binds to the macrophagelike cell line, J774… Show more

“…Cisplatin was applied using concentrations and an exposure time that cause accumulation in G2 phase of the cell cycle, followed by apoptotic cell death (Demarcq et al, 1994). Paclitaxel was used at a concentration (20 mM) in the range previously reported to stabilize microtubules (Manie et al, 1993;Shinohara-Gotoh et al, 1991) and inhibit the e ects of microtubule-disrupting agents (Manfredi et al, 1982). …”

“…Taxol appears to target microtubules specifically both in vitro and in vivo Horwitz, 1980, 1981;Manfredi et al, 1982;Manfredi and Horwitz, 1984;De Brabender et al, 1996). However, unlike classical antimicrotubule agents, such as colchicine and vinblastine, that induce microtubule disassembly and paracrystal formation (Wilson et al, 1974), the antitumour activity of taxol has been demonstrated to be associated with microtubule stabilisation (Manfredi and Horwitz, 1984;Horwitz et al, 1986) and with the arrest of cells in the G2/M phase of the cell cycle Slichenmyer and von Hoff, 1991;Rowinsky et al, 1992).…”

“…The taxanes had been identified as potential radiosensitizers when preclinical data revealed their lethal effects of inhibiting mitosis, interfering with the cell cycle, and encouraging apoptosis (Dorr et al, 1997;Kearns et al, 1997;Herscher et al, 1999;Parness et al, 1981;Manfredi et al, 1982). Taxane was shown to have significant activity in patients with metastatic esophageal cancer in the early 1990s (Ajani et al, 1994).…”
