Technical Note: Using enzyme-linked immunosorbent assays to evaluate humoral responses to vaccination against respiratory viruses in beef cattle

Abstract: This experiment evaluated humoral responses in beef calves vaccinated against parainfluenza-3 virus (PI3), bovine respiratory syncytial virus (BRSV), and bovine herpesvirus-1 (BHV-1) using via serum neutralization (SN) tests or enzyme-linked immunosorbent assays (ELISA). Blood samples were collected from 50 overtly healthy Angus-influenced steers (183 ± 3 kg of body weight, 212 ± 2 d of age) on d 0, 21, 35, and 49 of the experiment. Steers were vaccinated against respiratory viruses on d 0 and 21. Blood was pr… Show more

“… b Heifers received vaccination against respiratory pathogens on d 0 (Vista Once SQ; Merck Animal Health, Madison, NJ). Samples collected on d 0, d 13, d 27, d 41, and d 55 were analyzed and results expressed as sample:positive control ratio (%) as in Cooke et al (2020) . Results from d 0 was used as covariate in each respective analysis.…”

“…There were a few apparent divergences in the conclusions relating to serology investigations, ELISA vs VNT: ELISA methods are very frequently used in field studies, for convenience and cost reasons, in preference to VNT determinations. Although it is possible to demonstrate a good correlation between ELISA and VNT (Cooke et al 2020), VNT remains the 'gold standard' when there are discrepancies, because it is considered a biologically more relevant test.…”

“…Therefore, in a sense, the detection of false-positive and falsenegative pathogenic antibodies in some sera can not be avoided. Indeed, false positives and false negatives have been reported in the detection of serum antibodies by ELISA [19]. On the other hand, this may be related to the low titer of pathogenic antibodies in the serum.…”
