Telomeres in the mouse have large inter-chromosomal variations in the number of T 2 AG 3  repeats

Abstract: The ultra-long telomeres that have been observed in mice are not in accordance with the concept that critical telomere shortening is related to aging and immortalization. Here, we have used quantitative f luorescence in situ hybridization to estimate (T 2 AG 3 ) n lengths of individual telomeres in various mouse strains. Telomere lengths were very heterogeneous, but specific chromosomes of bone marrow cells and skin fibroblasts from individual mice had similar telomere lengths. We estimate that the shortest te… Show more

“…Telomerase knockout mouse exhibit decreases fertility, reduced litter size in subsequent generations and eventual sterility [9]. Hetrogenetiy of mouse and human somatic cell telomere length is also seen in oocytes [47]. Other possible mechanisms such as loss of telomerase activity due to mutation in the gene cannot be ruled out.…”

“…To demonstrate colocalization of GFP-tagged proteins with telomeric DNA, we analyzed HeLa cells transiently expressing CFP-POT1 and YFP-TRF2 with subsequent fluorescence in situ hybridization by using a Cy3-coupled PNA probe for telomeric DNA (39). These studies clearly demonstrate colocalization of tagged hPOT1 and TRF2 proteins with telomeric DNA (Fig.…”

“…Immunofluorescence images were obtained by using a Leica DMRXA microscope equipped with a Sensys charge-coupled device camera (Photometrics) and QFluoro software (Leica). To demonstrate colocalization of GFPtagged proteins with telomeric DNA fluorescence in situ hybridization with a Cy3-coupled PNA probe for telomeric DNA was performed (39). For each GFP fusion protein, three independent clones with relatively low GFP fusion protein expression level were selected for bleaching experiments.…”

“…Telomere fluorescence was expressed as numbers of molecules of equivalent soluble fluorochrome. Quantitative fluorescence in situ hybridization (Q-FISH) of activated splenocytes and MEFs was performed as described previously (14,78). A Cy3-labeled (CCCTAA) 3 peptide nucleic acid (Applied Biosystems) was used as a probe.…”
