TGF-β Signal Transduction in Oro-facial Health and Non-malignant Disease (Part I)

Abstract: ABSTRACT:The transforming growth factor-beta (TGF-␤) family of cytokines consists of multi-functional polypeptides that regulate a variety of cell processes, including proliferation, differentiation, apoptosis, extracellular matrix elaboration, angiogenesis, and immune suppression, among others. In so doing, TGF-␤ plays a key role in the control of cell behavior in both health and disease. In this report, we review what is known about the mechanisms of activation of the peptide, together with details of TGF-␤ … Show more

“…Although this growth factor pool may be used by the depositing cells themselves (10,11), it is possible that it also serves as a message for subsequent developmental processes governing the epithelial closure. Intriguingly, TGF-␤/BMP is the same signaling pathway associated with scar formation and scarless wound healing (12)(13)(14), and TGF-␤3 was demonstrated to mediate antiscarring effects. Important implications of TGF-␤ signaling (see Ref.…”

“…41 TGF-b1 is one of the key cytokines with pleiotropic properties that has both proinflammatory and anti-inflammatory features in the regulation of the inflammatory infiltrate. This cytokine is a chemoattractant for neutrophils, monocytes, mast cells and lymphocytes.…”

“…Our results confirm that increased gingival inflammation is associated with high levels of TGF-isoforms. It is well known that TGF-is one of several cytokines able to regulate inflammation and immune responses and the fact that TGF-mediates leukocyte recruitment, adhesion and activation suggests that it play a key role in the host response to bacterial and immunological insults (Prime et al, 2004), even there is not a clear evidence for its role in the pathogenesis of the periodontal disease. But TGF-has also a marked effect on ECM homeostasis, being an important mediator of fibroblast proliferation and ECM synthesis.…”
