The 60‐kDa precursor to the dithiothreitol‐sensitive tetrameric protease of spinach thylakoids: structural similarities between the protease and polyphenol oxidase

Abstract: The 60-kDa precursor to the 39-kDa dithiothreitolsensitive protease was purified from photosystem II membranes of spinach. When partially purified 60-kDa protein was stored at 4°C, the protein was degraded to fragments of 39 and 21 kDa. The 39-kDa fragment was suggested to be identical to the 39-kDa protease from effects of dithiothreitol on these polypeptides. The N-terminal amino acid sequences of the 60-kDa protein and the 39-kDa protease were the same, APILPDVEK-, suggesting that the latter was derived fro… Show more

“…A curious finding on the possible ability of PPO to act as a protease has been reported (Sokolenko et al, 1995;Kuwabara, 1995;Kuwabara et al, 1997;Kuwabara and Katoh, 1990). According to these reports a protein said to be identical with PPO in structure and properties can under certain conditions oxidatively degrade a low molecular weight protein located in chloroplasts.…”

“…In addition, it includes two isoforms of a plastid polyphenol oxidase (1748 and 1754) that potentially regulates photosynthesis (Dry and Robinson, 1994;Kuwabara, 1995). Another transcript linked to chloroplast metabolism encodes a plastid plastocyanin (TC65998) whose role may be to facilitate electron transport between the cytochrome b 6 f complex and PSI (Sugawara et al, 1999).…”

“…Spinach thylakoid PPO has also been correlated with dithiothreitol-sensitive proteases (39 and 60 kDa) which were extracted with 1 M NaCl from spinach PSII membranes (Kuwabara & Hashimoto, 1990;Kuwabara, 1995). These proteins had the same N-terminal sequence, APILPDVEK-, as the above-mentioned 64 kDa protein.…”

“…At present 27 polypeptides are identified as either PSII subunits or proteins associated with the PSII complex (Andersson & Styring, 1991;Irrgang et al, 1993;Kuwabara & Hashimoto, 1990;Kuwabara, 1995;Zhang et al, 1995). Only two of the 27 proteins, the heterodimer of the D1 and D2 subunits which forms the reaction center, bind all of the redox components that are necessary for primary charge separation and water oxidation.…”
