There is a need for more efficient and eco-friendly approaches to overcome increasing microbial infections. Bacteriocins and chitinases from Bacillus spp. can be powerful alternatives to conventional antibiotics and antifungal drugs, respectively. The purpose of this study was to assess the inhibitory potential of bacteriocins and chitinase enzymes against multiple resistant bacterial and fungal pathogens. Bacterial isolates were selected by growth on minimal salts medium and after that were morphologically and biochemically characterized. The physiochemical characterization of bacteriocins was carried out. The inhibitory potential of bacteriocins towards six pathogenic bacteria was determined by the well diffusion assay while chitinase activity towards three fungal strains was determined by the dual plate culture assay. Two bacterial strains (WW2P1 and WRE4P2), out of nine showed inhibition of K. pneumonia, P. aeruginosa, E. coli and MRSA while WW4P2 was positive against S. typhimurium and E. coli and WRE10P2 against P. aeruginosa, S. pneumoniae. Two bacterial isolates (WW3P1 and WRE10P2) were chosen for further study on the basis of their antifungal activities. Of these, WW3P1 isolate was more effective against A. fumigatus as well as A. niger. The proteinaceous nature of the bacteriocins was confirmed by treatment of the crude extract with proteinase K. It was found that the inhibitory activity of strain WW3P1 against E. coli was highest at 20 °C, and against S. pneumoniae it was at 20 °C and pH 10 after treatment with EDTA. Inhibition by strain the WRE10P2 against P. aeruginosa was highest at 20 °C and pH 14. It was found that EDTA increased the inhibitory activity of strain WW2P1 against P. aeruginosa, K. pneumoniae and E. coli by 2 ± 0.235, 3.5 ± 0.288, 2.5 ± 1.040 times, respectively, of strain WRE4P2 against P. aeruginosa and E. coli by 2.5 ± 0.763, 2.7 ± 0.5 times, respectively, and of strain WRE10P2 against S. pneumoniae by 3 ± 0.6236 times. The isolates have promising inhibitory activity, which should be further analyzed for the commercial production of antimicrobials. Article highlights The current study aimed to isolate the microbiome from wheat plant (Triticum aestivum L.), to screen for bacteriocin production and to assess its antimicrobial activity against human pathogens. Forty-one phenotypically different bacterial colonies were subjected to bacteriocin purification from which 25 colonies showed positive reactions. These 25 bacterial isolates were screened against six different human bacterial pathogens using the well diffusion method to check the antimicrobial activity. Out of nine bacterial isolates, WW3P1 and WRE10P2 were able to degrade the chitin and utilize it as their sole energy source. Strain WRE4P2 exhibited partial inactivation in its activity against MRSA after treatment with proteinase K.

Penyakit antraknosa merupakan penyakit utama pada tanaman cabai yang dapat menyebabkan kegagalan panen dan kerugian mencapai 80 %. Tujuan penelitian untuk menilai perkembangan penyakit antraknosa cabai di Kecamatan Baturraden, menguji pengaruh komponen epideminya terhadap pertumbuhan jamur Colletotrichum gloeosporioides dan penekanan penyakit pada buah cabai. Metode penelitian yang digunakan adalah survey dengan pengambilan sampel secara purposive random sampling di empat desa di Kecamatan Baturraden Kabupaten Banyumas. Pengujian pengaruh komponen epidemi dilakukan di Laboratorium Perlindungan Tanaman Faperta Unsoed dengan menumbuhkan jamur C. gloeosporioides pada beberapa suhu dan menguji penekanan penyakit pada buah cabai. Rancangan yang digunakan adalah Rancangan Acak Lengkap. Variabel pengamatan yaitu intensitas penyakit, laju infeksi, kecepatan pertumbuhan jamur, persentase penghambatan pertumbuhan jamur dan penekanan penyakit pada buah cabai. Hasil penelitian menunjukkan bahwa penyakit antraknosa di desa Kemutug Lor menunjukkan intensitas penyakit tertinggi yaitu 76% dengan laju infeksi 0,345 unit/hari. Suhu optimum yang mendukung pertumbuhan C. gloeosporioides yaitu 29oC, dengan kecepatan pertumbuhan 14,72 mm.hari-1. Pertumbuhan C. gloeosporioides dihambat oleh bakteri endofit cabai BE2 sebesar 78,6%. Bakteri endofit cabai dapat menekan penyakit antraknosa pada buah cabai dengan efektivitas 30,93%. Anthracnose is the main disease in chili that can cause crop failure and losses up to 80%. The aim of the study was to assess the development of chili anthracnose in the Baturraden district, to examine the effect of its epidemic components on the growth of Colletotrichum gloeosporioides, and suppression of anthracnose. The research method used was a survey with purposive random sampling in four villages in Baturraden district, Banyumas Regency. Testing the effect of epidemic components was carried out in Plant Protection Laboratory, Faculty of Agriculture Unsoed by growing the C. gloeosporioides at several temperatures, and testing the disease suppression of chilies with chili endophytic bacteria. The design used was a Completely Randomized Design. The variables observed were disease intensity, infection rate, fungal growth rate, percentage of inhibition of fungal growth, and disease suppression in chilies. The results showed that chili anthracnose in the village of Kemutug Lor showed the highest intensity of 76% with an infection rate of 0,345 units per day. The optimum temperature that supported the growth of C. gloeosporioides was 29oC with a growth rate of 14,72 mm day-1. Growth of C. gloeosporioides was inhibited by endophytic bacteria BE2 chili by 78,6%. Chili endophytic bacteria could suppress anthracnose in chilies by 30,93% effectivity.

The main disease in chili is fusarium wilt caused by Fusarium oxysporum f. sp. capsici that can cause losses up to 100%. The aim of this study was to obtain a solid formula for the endophytic bacterium Bacillus cereus strain SLBE3.1AP with a carrier that can be stored longer and is effective for controlling Fusarium oxysporum f. sp. capsici. This research was an experimental study to see the ability of the solid formula of Bacillus cereus bacteria strain SLBE3.1AP in controlling Fusarium oxysporum f. sp. capsici using Completely Randomized Design consisting of 15 treatments and 3 replications. The treatments consisted of carrier material for the formula bagasse, rice straw, bran, fungicide and control. Each solid formula of Bacillus cereus strain SLBE3.1.AP was introduced to chili seeds and seedlings. The results showed that the best formula for controlling Fusarium oxysporum f.sp. capsici and increasing the growth of chili plants was a solid formula of B. cereus strain SLBE3.1AP with 6 weeks storage of bagasse, 4 weeks of storage of rice straw, and 6 weeks of storage of rice bran.
