The actions of calcitonin gene related peptide and vasoactive intestinal peptide as vasodilators in man in vivo and in vitro.

Abstract: 1 The two peptides calcitonin gene related peptide (CGRP) and vasoactive intestinal peptide (VIP) produced marked dilatation of the forearm vascular bed when infused via the brachial artery. 2 CGRP relaxed preconstricted segments of human radial, coronary, gastric and cerebral arteries in an endothelium dependent manner. 3 VIP relaxed human gastric and transverse cervical arteries in an endothelium dependent manner, but relaxation of the human pulmonary artery was not dependent on endothelium. 4 The character… Show more

“…In rings of rat aorta and mesenteric artery, human a-CGRP acts in an endothelial dependent manner like acetylcholine (Brain et al, 1985;Kubota et al, 1985;Al-Kazwini et al, 1987). In human radial, coronary and gastric arteries the relaxation evoked by human a-CGRP was also endothelium dependent (Thom et al, 1987). In contrast to these observations, in rings of cat middle cerebral artery the response was independent of the endothelium (Hanko et al, 1985).…”

“…In the original description of its cardiovascular activity (Brain et al, 1985), vasodilatation induced by CGRP in the rat aorta was endothelium-dependent, and similar findings were obtained with human vessels (Thom et al, 1987). However, in the bovine coronary artery vasodilatation in response to CGRP did not require the presence of endothelium (Greenberg et al, 1987).…”

“…The vasodilator effect of calcitonin gene-related peptide (CGRP) (Amara et al, 1982) has been extensively studied in the coronary circulation of several mammals Greenberg et al, 1987;Shoji et al, 1987;Franco-Cereceda, 1988;Beny et al, 1989) including man (McEwan et al, 1986;Thom et al, 1987). CGRP-immunoreactivity is present in high concentration in cardiac tissue and in nerve fibres associated with coronary blood vessels (Mulderry et al, 1985).…”
