The Anti-Proliferation Activity and Mechanism of Action of K12[V18O42(H2O)]∙6H2O on Breast Cancer Cell Lines

Abstract: Polyoxometalates (POMs) are inorganic clusters that possess potential anti-bacterial, anti-viral, and anti-tumor activities. Herein, the in vitro anti-proliferation activities of K12[V18O42(H2O)]∙6H2O (V18) have been investigated on the MCF-7 and MDA-MB-231 cell lines. The results indicated that V18 could inhibit the proliferation of MCF-7 (IC50, 11.95 μM at 48 h) in a dose-dependent manner compared to the positive control, 5-fluorouracil (5-Fu, p < 0.05). The anti-proliferation activity of V18 might be mediat… Show more

“…These present various combinations of V IV and V V centres, [36] from which the value of the charge nÀ , the structural features and the magnetic, [37] catalytic, [49,50] and biological properties depend. [46,51] These POVs are able to incorporate in the centre of the cage various types of anions such as Cl À , Br À , I À , HS À , HCOO À , CO 3 2À , N 3 À , NO 2 À , NO 3 À or, also, neutral species like water. To the best of our knowledge, the cage observed in our structure is the second one with composition V 18 O 46 , after [V V 16 V IV 2 O 46 (NO 3 )] 5À .…”

“…Instead, in literature, five types of octadecavanadate (V 18 ) cages with composition [V 18 O 39 ] n− , [34] [V 18 O 41 ] n− , [35] [V 18 O 42 ] n− , [36–45] [V 18 O 44 ] n− , [46,47] and [V 18 O 46 ] n− , [48] have been reported. These present various combinations of V IV and V V centres, [36] from which the value of the charge n−, the structural features and the magnetic, [37] catalytic, [49,50] and biological properties depend [46,51] . These POVs are able to incorporate in the centre of the cage various types of anions such as Cl − , Br − , I − , HS − , HCOO − , CO 3 2− , N 3 − , NO 2 − , NO 3 − or, also, neutral species like water.…”

“…The anti-proliferation activity of another POV, V 18 , was observed to affect the cellular cycle, and to mediate the arrest of MCF-7 cells in the G2/M phase and induction of apoptosis, besides DNA-, BSA-, and HSA-binding [85]. POV studies were also performed with U-87 and human liver SMMC-7721 cancer cells, and cell cycle arrest, DNA damage, and apoptosis were observed [85,94].…”

“…Similarly, POT and POMos studies together represent the majority (90%), whereas for POVs and PONbs, lower percentages can be found. In fact, more than 120 articles/papers have been published with POMos and polyoxotungstates POTs in antitumor studies [8,[10][11][12][13][80][81][82][83][84][85][86][87][88][89]. POMs in cancer therapy and diagnostics, their modes of action, and future perspectives were reviewed [2,[90][91][92][93].…”

“…Mixed-valence polyoxovanadates (MVPs) have contributed to an increase in the number of potential therapeutic agents for metabolic diseases and cancer [ 25 , 26 , 27 , 28 ]. The MVP [(Me 4 N) 6 [V 15 O 36 Cl] is a polyoxovanadate (abbreviated V 15 , see Figure 1 ) that contains six Me 4 N + counterions, and the vanadium-oxygen cage of V 15 consists of one central chlorine atom, eight vanadium(IV) and seven vanadium(V) atoms, and 36 oxygen atoms ( Figure 1 ).…”
