The anti-tumor effect of ursolic acid on papillary thyroid carcinoma via suppressing Fibronectin-1

Abstract: This study aims to discover the effects of ursolic acid (UA) on papillary thyroid carcinoma (PTC). Human PTC cells were under UA treatment, and cell viability, clone formation, and apoptosis were measured by MTT assay, clone formation assay, and flow cytometry, respectively. Expressions of apoptosis- and epithelial–mesenchymal transition (EMT)-related markers were determined via qRT-PCR and western blot. Fibronectin-1 (FN1) expression in thyroid carcinoma was analyzed by GEPIA2 and qRT-PCR. The effects of over… Show more

“…Sponziello et al con rmed that the proliferation, adhesion, migration, as well as invasion of PTC cells were signi cantly suppressed after the silencing of FN1 [25]. Ursolic acid inhibited PTC cell viability, proliferation, and EMT but promoted apoptosis via suppressing FN1, thus exert antitumor effects in vivo [26]. As a calcium-dependent phospholipid-binding protein, ANXA1(Annexin A1) presents a molecular weight of 37 kDa and is involved in cell growth, differentiation, and apoptosis [27,28].…”

“…A number of other studies have demonstrated that ursolic acid has anti-cancer effects against various cancer types including esophageal cancer [108], pancreatic cancer [109], and colorectal cancer [110] via a number of different mechanisms, including the induction of ROS-mediated autophagy [108] and ER stressmediated RAGE inhibition, promoting apoptosis and autophagy [109], and up-regulation of ROS and caspase-3, -8, and -9, thereby inducing apoptosis [92]. Further anti-tumor effects of ursolic acid have been observed in papillary thyroid carcinoma cells, mediated via enhancement of fibronectin-1 expression and subsequent induction of apoptosis [111].…”

“…Ursolic acid (3‐hydroxy‐12‐ursen‐28‐oic acid, UA, Figure 1) belongs to pentacyclic triterpenoids that mainly exist in plants such as the whole grass of Prunella vulgaris, and Ilex. It exhibits wide range of pharmacological activities, including anti‐oxidation (Wang et al, 2018), anti‐bacterial (Qian et al, 2020), anti‐ulcer (Liu et al, 2016), anti‐inflammatory (Wang et al, 2018), anti‐tumor (Cao et al, 2020) anti leukemia (da Silva et al, 2021), lowering blood glucose (Alkreathy & Ahmad, 2020), as well as obvious stability and cooling effects on the central nervous system (Colla et al, 2021), and liver protection (Ma et al, 2021). In recent years, it has attracted much attention due to its strong pharmacological activity, slight side effects, and a vast range of sources.…”
