The Application of the CRISPR-Cas System in Antibiotic Resistance

Abstract: The emergence and global epidemic of antimicrobial resistance (AMR) poses a serious threat to global public health in recent years. AMR genes are shared between bacterial pathogens mainly via horizontal gene transfer (HGT) on mobile genetic elements (MGEs), thereby accelerating the spread of antimicrobial resistance (AMR) and increasing the burden of drug resistance. There is an urgent need to develop new strategies to control bacterial infections and the spread of antimicrobial resistance. The clustered regul… Show more

“…Mobile genetic elements, such as insert sequences (ISs), transposons (Tns) and integrons, play an important role in increasing antibiotic resistance ( Partridge et al, 2018 ). Bacteria share MGEs and their associated resistance genes with other bacterial species via horizontal gene transfer (HGT), which has promoted the accumulation and dissemination of antibiotic resistance genes (ARGs) in bacteria ( Tao et al, 2022 ). Integrons constitute an important and near-ubiquitous class of genetic elements.…”

“…The emergence of drug-resistant bacteria has greatly reduced the efficacy of current antibiotics, resulting in increased morbidity, mortality, and healthcare costs. To address this challenge, underexploited targets such as MurA have been identified as promising alternatives to novel antibacterial methods, such as the CRISPR-Cas system . Covalent inhibition of MurA, for example, with the clinically used fosfomycin, is a validated strategy for antibacterial drug development.…”

“…Type III CRISPR mechanisms demonstrated indiscriminate targeting for DNA cleavage, unlike the Type II CRISPR system, which relies on the Cas9 nuclease to cut specific DNA sequences . Therefore, the rational design of CRISPR-based tools like novel antibiotics has shown strong inclinations toward using Cas9 nucleases . However, the heavy reliance on these specific machinery raises concerns about future resistance mechanisms.…”
