The Association of Gastric Cancer Risk with Plasma Folate, Cobalamin, and Methylenetetrahydrofolate Reductase Polymorphisms in the European Prospective Investigation into Cancer and Nutrition

Abstract: Previous studies have shown inconsistent associations of folate intake and polymorphisms of the methylenetetrahydrofolate reductase (MTHFR) gene with gastric cancer risk. Our nested case-control study within the European Prospective Investigation into Cancer and Nutrition cohort is the first prospective study of blood folate levels and gastric cancer. Gastric cancer cases (n = 247) and controls (n = 631) were matched for study center, age, sex, and time of blood donation. Two common single nucleotide polymorph… Show more

“…In a Mexican population, a significant reduction in gastric cancer risk was observed among individuals with high folate intake and who carried the MTHFR 677TT genotype (Galvan-Portillo et al, 2009). Two other studies detected a non-significant protective effect on gastric cancer risk (Zuniga-Noriega et al, 2007;Vollset et al, 2007). Here, we observed that MTHFR 677TT was associated with a reduced risk of gastric cancer, and the MTHFR 677T allele was associated with a marginally decreased risk.…”

“…Studies conducted in China (Qin et al, 2008), Iran (Saberi et al, 2012), Korea (Cui et al, 2010), Mexico (ZunigaNoriega et al, 2007;Galvan-Portillo et al, 2009), Norway (Vollset et al, 2007), and Denmark (Zacho et al, 2011) identified a significant association of the MTHFR C677T polymorphism with gastric cancer. Studies in China, Iran, and Denmark indicated a significantly increased risk of gastric cancer, with ORs ranging from 1.4-2.6 (Qin et al, 2008;Zacho et al, 2011;Saberi et al, 2012).…”

“…Inverse association of cobalamin to gastric cancer occurrence also exists, while MMA (which is elevated under vitamin B12 deiciency) is positively associated with gastric cancer [50]. This result could be due to worsen vitamin B12 status in atrophic gastritis that often precedes gastric cancer.…”

“…Several studies have reported that a homozygous variant genotype of the polymorphism of MTHFR C677T was associated with an increased risk of CRC. Certain studies have reported that individuals with the MTHFR 677TT genotype had a decreased risk of CRC (8), whereas other authors observed no association between the MTHFR C677T genotype and genetic susceptibility to CRC (9)(10)(11). Considering the number of studies performed thus far, inconsistent results have been reported on the association of MTHFR C677T gene polymorphisms with genetic susceptibility to CRC.…”
