The biological relevance of PCSK9: when less is better…

Abstract: Proprotein Convertase Subtilisin/Kexin-type 9 (PCSK9) is a circulating negative regulator of hepatic low-density lipoprotein receptor (LDLR) which clears cholesterol from blood. Gain-of-function genetic mutations which amplify PCSK9 activity have been found to cause potentially lethal familial hypercholesterolemia. Inversely, reduction of its activity through loss-of-function genetics or with pharmaceuticals was shown to increase hepatic LDLR, to lower blood cholesterol, and to protect against cardiovascular d… Show more

“…86 A significant correlation has been found between PCSK9 and glucose metabolism, 87 but studies linking PCSK9 LOF variants to the risk of developing diabetes are conflicting. 88,89 PCSK9 may also be involved in the pathogenesis of fatty liver disease. For example, PCSK9 rs11591147 (p.R46L) LOF has a protective effect on hepatic steatosis, NAFLD/NASH, and fibrosis.…”

“…Indeed, Perrone et al reported the prognostic role of hypercholesterolemia in terms of overall survival and non-progression-free survival in patients treated with immune checkpoint inhibitors [49]. Recently, a research focus has been on proprotein convertase subtilisin/kexin-type 9 (PCSK9), a circulating negative regulator of LDL receptors that reduces blood Chol [50]. Genetic mutations responsible for amplification of PCSK9 activity are considered a potential cause of lethal familial hypercholesterolemia.…”
