The brain angiotensin IV/AT4 receptor system as a new target for the treatment of Alzheimer's disease

Abstract: The brain renin-angiotensin system (RAS) regulates several physiologies including blood pressure, body sodium and water balance, cyclicity of reproductive hormones and related sexual behaviors, and the release of pituitary gland hormones. These physiologies are under the control of the angiotensin II (AngII)/AT 1 receptor subtype system. The AngII/AT 2 receptor subtype system is expressed during fetal development and is less abundant in the adult. This system appears to oppose growth responses facilitated by a… Show more

“…In their study, Hallberg and co-workers designed cyclic analogs of the hexapeptide angiotensin IV (129) [172], which is known to play important roles in cognition processes, like memory and learning [173]. Angiotensin IV binds with high affinity to the insulin-regulated aminopeptidase (IRAP), an enzyme localized in the brain Synthesis of Cyclic Peptides and Peptidomimetics by Metathesis Reactions and is associated with the mentioned mental processes.…”

“…We determined that Nle 1 -AngIV improved memory in animal models of AD. This nootropic effect derived from its N-terminal region given that tetra-and tripeptide fragments of Nle 1 -AngIV were also capable of overcoming scopolamine-induced amnesia [70][71][72] . In addition, Nle 1 -AngIV and these shorter fragments, facilitated hippocampal synaptic connectivity via synaptogenesis [70] .…”
