The Chemical Biology of Protein Phosphorylation

Abstract: The explosion of scientific interest in protein kinase-mediated signaling networks has led to the infusion of new chemical methods and their applications related to the analysis of phosphorylation pathways. We highlight some of these chemical biology approaches across three areas. First, we discuss the development of chemical tools to modulate the activity of protein kinases to explore kinase mechanisms and their contributions to phosphorylation events and cellular processes. Second, we describe chemical techn… Show more

“…In a typical phosphorylation event, a protein kinase transfers the γ-phosphate from ATP to specific amino acids of a protein to initiate signal transduction; in eukaryotes, these phosphorylated amino acids are usually Ser, Thr, and Tyr residues (3,4). In contrast, His/Asp-based phospho-relay of two-component systems has been well recognized as a classic mode of signal transduction in the bacterial world (5).…”

“…The performance of NiZnFe 2 O 4 has also been compared with other approaches including TiO 2 27-30 , Fe 3 þ -IMAC 31 and HAP (hydroxyapatite) 32,33 (Supplementary Note 2; Supplementary Table S2). These materials can bind with both mono-and multi-phosphorylated peptides without selectivity ( Supplementary Fig.…”

“…R eversible protein phosphorylation is an important regulatory mechanism for prokaryotic and eukaryotic cells 1,2 . Cycles of phosphorylation and dephosphorylation cause conformational changes in structures of many enzymes and receptors and also function as electrostatic switches to modulate the on and off states of many signalling pathways [3][4][5] .…”

“…[80][81][82] If one embraces Julian's idea of antibiotics as mediators of cell-cell communication, it is interesting also to consider aminoglycoside phosphotransferases potentially serving as modulators of such signaling pathways-much as kinases that act on key regulatory proteins can profoundly alter cellular and developmental signaling networks. [83][84][85][86] Julian's challenge to us, to rethink just what roles antibiotics may play in Nature, illustrates yet once again his creativity, his clarity of thought, his biochemical and evolutionary insights, and why it was such an immense pleasure and privilege to work with him in Geneva and Madison some four decades ago.…”
