“…These data indicated that RprY was acetylated in vivo with the involvement of Pat activity. Several studies showed that proteins can be chemically acetylated with either acetyl CoA or acetyl phosphate in vitro [2123]. To separate protein acetylation from phosphorylation functions, we chose to examine acetyl CoA as acetyl donor.…”

“…Western blotting using anti-acetyl-lysine antibodies and protein visualization by Coomassie staining showed that acetylation of RprY was dependent on the concentration of acetyl-CoA and the length of incubation (Figure 4(b,c)). These properties, including retention of the modification after SDS-PAGE, suggest that RprY is autoacetylated covalently [21] and specifically, since Pat itself is not acetylated under the same conditions (Figure 5(a)), but does appear to enhance the acetylation of RprY in the presence of acetyl CoA (Figure 5(b,c)). The CobB deacetylase recognized autoacetylated RprY as a substrate, and in the presence of cofactor NAD + removed the modification (Figure 5(d)).…”

“…The data mining approach is shown in Figure 1 Table 2. 18 Results (peptide sequences, modifications, match scores, etc) were exported to Excel,19 where sequences were filtered to retain only the top scoring match for each spectrum with 20 an expectation value ≤ 0.05. This corresponds to a match score at the 95% confidence 21 interval of the Mascot scoring scheme for a given search.…”

“…Such a study highlights the importance of 18 minimizing chemical side reactions so that the biological modifications are not masked. 19 Many modifications, such as methylations and certain oxidations/hydroxylations, 20 are more likely to be post translational modifications rather than in vitro side-reactions. 21 The most extensive of these reactions identified in Dataset 1 were hydroxyproline (34 22 peptides), methylated D/E (69 peptides), dimethylated K (21 peptides), and a +42 Da 23 modification on K (18 peptides) ( Table 1).…”
