The Circadian Biology of Heart Failure

Abstract: Driven by autonomous molecular clocks that are synchronized by a master pacemaker in the suprachiasmatic nucleus, cardiac physiology fluctuates in diurnal rhythms that can be partly or entirely circadian. Cardiac contractility, metabolism, and electrophysiology, all have diurnal rhythms, as does the neurohumoral control of cardiac and kidney function. In this review, we discuss the evidence that circadian biology regulates cardiac function, how molecular clocks may relate to the pathogenesis of heart failure, … Show more

“…In the cardiovascular system, the circadian clock regulates several key mechanisms such as blood pressure (Costello & Gumz, 2021), heart rate (El Jamal et al, 2023), and cardiac contractility (Latimer & Young, 2022). BMAL1 expression is crucial for mitochondrial activities and cardiac function, and its deletion in human embryonic stem F I G U R E 2 Alterations in the core-clock machinery induce expression variations in clock-regulated genes associated with aging on Earth and increased risk of health problems due to spaceflight.…”

“…4 This reflects behavior, environmental rhythms, and endogenous circadian variation in physiological mechanisms from blood clotting 5 to cardiac arrhythmias. 6 However, although there has been extensive study of diurnal variation (ie, the nonlinear combination of endogenous circadian and evoked changes from sleep/wake state, activity levels, posture, eating, and other factors) in systemic hemodynamics, 7,8 there has been limited focus on diurnal variation in cerebral blood flow (CBF) and its dependence on both systemic physiology and cerebrovascular regulatory mechanisms.…”
