The class III phosphatidylinositol 3-kinase Vps34 in Saccharomyces cerevisiae

Abstract: Abstract:The class III phosphatidylinositol 3-kinase Vps34 (vacuolar protein sorting 34) catalyzes for the formation of the signaling lipid phosphatidylinositol-3-phopsphate, which is a central factor in the regulation of autophagy, endocytic trafficking and vesicular transport. In this article, we discuss the functional role of the lipid kinase Vps34 in Saccharomyces cerevisiae.

“…Analysis of the Atg1-dependent phosphoproteome revealed a similarly complex network of interactions specifically with the TORC1 signaling branch. For instance, Atg1 may feedback regulate TORC1 by (directly or indirectly) (1) impinging on Seh1 and Sea4, two subunits of the SEACAT complex that controls TORC1 through the Rag GTPases (Panchaud et al, 2013); (2) regulating Ser 327 phosphorylation within the TORC1 subunit Tco89; and/or (3) controlling the Ser 445 /Ser 449 phosphorylation within the PI3-kinase Vps34 that is key for TORC1 and autophagy activation (Reidick et al, 2017;Tanigawa and Maeda, 2017) (Figure 3). Lastly, Atg1 also converges with TORC1 on Gcn2.…”

“…The catalytic subunit is VPS34, which only uses PtdIns as substrate in order to generate PtdIns3P [ 9 , 10 , 11 ]. Moreover, it is the only PI3K in yeast and plants [ 12 , 13 , 14 ].…”

“…The role of Gpa1 in protein and lipid recycling could be explained by a model presented by Dohlman and colleagues (Slessareva et al, 2006), whereby Gpa1 has a role outside of surface GPCR signalling and can functionally connect with the yeast phosphatidylinositol 3-kinase (Vps34/Vps15). As Vps34 / Vps15 activity produces phosphatidylinositol 3-phosphate (PtdIns 3-P), which is required for various post-Golgi membrane trafficking events (Reidick et al, 2017), we propose lipid regulation via Gpa1 attenuates recycling in response to glucose starvation. The finding that Gpa1 physically binds Gpa2 (Ho et al, 2002), suggests these observations could be functionally connected.…”
