The clinical significance of allergen-specific IgG4 in allergic diseases

Abstract: IgG4 is a subclass of IgG antibody with a unique molecular feature of (Fragment antigen- binding) Fab-arm exchange, allowing bispecific antigen binding in a mono-valent manner. With low binding affinity to C1q and Fcγreceptors, IgG4 is incapable of forming immune complexes and activating the complement pathway, exhibiting a non-inflammatory feature. IgG4 is produced similarly to IgE and is considered a modified reaction to IgE class-switching response under certain conditions. It could also counteract IgE-acti… Show more

“…As previously mentioned, the effectiveness of allergen‐specific immunotherapy (AIT) was evaluated by a reduction in circulating IgE levels and an increase in IgG4 expression in peripheral blood. Our research corroborated these findings at the transcriptional level 7,11,20 . Our study delineated the intricate landscape of immune cell populations during ICLIT, highlighting alterations in IGHE , IGHA , and IGHGs within memory B cell subsets.…”

“…Our research corroborated these findings at the transcriptional level. 7,11,20 Our study delineated the intricate landscape of immune cell populations during ICLIT, highlighting alterations in IGHE, IGHA, and IGHGs within memory B cell subsets. The rise of allergen-specific IgG2 and IgG4 through the intermediate allergen-specific IgG1+ memory B cells required CSR.…”

“…IgA in mucosal secretions can bind to allergens, forming immune complexes that prevent the allergens from crossing the mucosal barrier and entering the circulation (immune exclusion). IgA can neutralize allergens' biological activity by binding to them and preventing them from inducing an immune response 57,58 …”
