The Co-Culture of Staphylococcal Biofilm and Fibroblast Cell Line: The Correlation of Biological Phenomena with Metabolic NMR1 Footprint

Abstract: Staphylococcus aureus is one of the most prevalent pathogens associated with several types of biofilm-based infections, including infections of chronic wounds. Mature staphylococcal biofilm is extremely hard to eradicate from a wound and displays a high tendency to induce recurring infections. Therefore, in the present study, we aimed to investigate in vitro the interaction between S. aureus biofilm and fibroblast cells searching for metabolites that could be considered as potential biomarkers of critical colo… Show more

“…They highlighted how these complementary analyses provided information on intracellular and extracellular targets, such as protein, DNA and cell wall. Czajkowska et al [ 36 ] employed 1 H NMR extracellular footprinting analysis to investigate in vitro interactions between S. aureus biofilm and fibroblasts. However, all these studies were performed as single measurements.…”

“…The viability and proliferation of the microbial cells were evaluated using quantitative culturing on the appropriate solid agar plates (Müller-Hinton agar for bacteria and Sabouraud agar for yeast (Biomaxima, Lublin, Poland)) and tetrazolium test every 24 hours for 3 days. The 24 h co-culture of microbes and fibroblasts was chosen as a model for antiseptic application, similarly as we have shown it earlier ( Czajkowska et al., 2021 ). Such prepared biofilm grown on the fibroblast-containing BC carrier was exposed to 2 mL of P, G or M antiseptics for 1 hour of contact time.…”

“…Devoid of the skin's protective barrier and repeatedly flooded with nutrient-rich exudate, chronic wounds are attractive niches for microbial colonization and formation of biofilm [4] . The results of recent studies indicate that biofilm is the dominant form of microbial existence in wounds [5,6] . Biofilm is frequently defined based on in vitro observations.…”
