The combination of CASA kinetic parameters and fluorescein staining as a fertility tool in cryopreserved bull semen

Abstract: Prediction of male fertility with in vitro semen quality parameters remains a challenge for the bull industry. Fluorescein staining and computer-assisted semen analysis (CASA) ensure kinetically proper and functionally objective results to improve spermatological parameter control. Therefore, we aimed to examine the CASA kinetic parameters and fluorescein staining of cryopreserved bull semen and its relationship with fertility. A total of 90 frozen straws from 5 Simmental bulls were evaluated. After semen was … Show more

“…Total motility (%), progressive motility (%), curvilinear velocity (VCL, µm/s), average path velocity (VAP, µm/s), straight line velocity (VSL, µm/s), amplitude of lateral head displacement (ALH, µm), beat‐cross frequency (BCF, Hz), linearity (LIN) [(VSL / VCL) × 100], wobble (WOB) [(VAP / VCL) × 100], straightness (STR) [(VSL / VAP) × 100] and hyperactivation [(VCL / VSL) × ALH] were determined. Approximately 400 spermatozoa per sample were evaluated in five microscopic fields (Inanc et al., 2018).…”

“…As males mature, it can be difficult to predict the ability of spermatozoa to fertilize ova in vivo; that prediction is complicated by many factors including bull breed and sperm characteristics [23] [24]. Pregnancy rates have been associated with sperm hyperactivity [18], and sperm velocity [25]. Our prediction analyses found that velocity characteristics of fresh semen were the most informative predictors of sperm quality following cryopreservation; specifically, VAP was selected in half of the traits predicted.…”

“…It could lead to questioning as to whether chosen settings were extreme enough to detect any differences? Tested range values were selected to be consistent with the value variations generally observed in published works (Chaveiro et al, 2006;İnanç et al, 2018;Lenz et al, 2011). Some kinematic parameters (such as VSL) increased with the threshold value modifications, but this was not significant.…”
