Distribution of the Harmful Bloom-Forming Cyanobacterium, <i>Microcystis aeruginosa</i>, in 88 Freshwater Environments across Japan

Microcystis aeruginosa was quantitatively surveyed in 88 freshwater environments across Japan within 3 weeks in 2011. In order to clarify the distribution pattern of M. aeruginosa at the intra-species level, three major genotypes, which were defined by 16S-23S rRNA inter-transcribed-spacer (ITS) regions, were selectively detected using quantitative real-time PCR assays. Of the 68 sites at which the Microcystis intergenic-spacer region of the phycocyanin (IGS-PC) gene was detected, the M. aeruginosa morphotype-related genotype (MG1) dominated in 41 sites, followed by the non-toxic M. wesenbergii-related genotype (MG3). A correlation analysis showed that total nitrogen and phosphate positively correlated with the abundance of IGS-PC, which positively correlated with microcystin synthetase gene abundance. A redundancy analysis of genotype compositions showed that pH positively correlated with the dominance of MG3 and negatively correlated with MG1, i.e., both toxic and non-toxic genotypes. Our survey of Microcystis populations over a wide area revealed that MG1 is a dominant genotype in Japan.

In deep drinking water reservoir ecosystems, the dynamics and interactions of community compositions of phytoplankton and eukaryotes during the mixing periods are still unclear. Here, morphological characteristics combined with high-throughput DNA sequencing (HTS) were used to investigate the variations of phytoplankton and the eukaryotic community in a large canyon-shaped, stratified reservoir located at the Heihe River in Shaanxi Province for three months. The results showed that Bacillariophyta and Chlorophyta were the dominant taxa of the phytoplankton community, accounting for more than 97% of total phytoplankton abundance, which mainly consisted of Melosira sp., Cyclotella sp., and Chlorella sp., respectively. Illumina Miseq sequencing suggested that the biodiversity of eukaryotes increased over time and that species distribution was more even. Arthropoda (6.63% to 79.19%), Ochrophyta (5.60% to 35.16%), Ciliophora (1.81% to 10.93%) and Cryptomonadales (0.25% to 11.48%) were the keystone taxa in common, contributing over 50% of the total eukaryotic community. Cryptomycota as a unique fungus was observed to possess significant synchronization with algal density, reaching a maximum of 10.70% in December (when the algal density distinctly decreased) and suggesting that it might affect the growth of algae through parasitism. Co-occurrence network patterns revealed the complicated and diverse interactions between eukaryotes and phytoplankton, suggesting that eukaryotes respond to variations in dynamic structure of the phytoplankton community, although there might be antagonistic or mutualistic interactions between them. Redundancy analysis (RDA) results showed that environmental variables collectively explained a 96.7% variance of phytoplankton and 96.3% variance of eukaryotic microorganisms, indicating that the temporal variations of phytoplankton and eukaryotic microorganisms were significantly affected by environmental conditions. This study shows that potential interactions exist between phytoplankton and eukaryotic microorganism communities, andcould improve our understanding of the ecological roles of phytoplankton and eukaryotic microorganisms in changing aquatic ecosystems. However, long-term investigations are necessary in order to obtain comprehensive understandings of their complicated associations.

Summary Toxic Microcystis spp. blooms constitute a serious threat to water quality worldwide. Aeromonas veronii was isolated from Microcystis sp. colonies collected in Lake Kinneret. Spent Aeromonas media inhibits the growth of Microcystis aeruginosa MGK isolated from Lake Kinneret. The inhibition was much stronger when Aeromonas growth medium contained spent media from MGK suggesting that Aeromonas recognized its presence and produced secondary metabolites that inhibit Microcystis growth. Fractionations of the crude extract and analyses of the active fractions identified several secondary metabolites including lumichrome in Aeromonas media. Application of lumichrome at concentrations as low as 4 nM severely inhibited Microcystis growth. Inactivation of aviH in the lumichrome biosynthetic pathway altered the lumichrome level in Aeromonas and the extent of MGK growth inhibition. Conversely, the initial lag in Aeromonas growth was significantly longer when provided with Microcystis spent media but Aeromonas was able to resume normal growth. The longer was pre‐exposure to Microcystis spent media the shorter was the lag phase in Aeromonas growth indicating the presence of, and acclimation to, secondary MGK metabolite(s) the nature of which was not revealed. Our study may help to control toxic Microcystis blooms taking advantage of chemical languages used in the interspecies communication.
